fzy5032/fzy1 mutant adults possess all appendages but have a dramatically reduced neuropil compared to controls. They exhibit poor motor function.

fzy5032/fzy1 larval brains show progressive neuroblast loss.

fzy5032/fzy1 mutant exhibit morphological and molecular marker changes consistent with aberrant onset of necrosis: in contrast to wild type, neuroblasts in fzy5032/fzy1 mutant brains often do not have identifiable nuclei, and their cytoplasm appears largely devoid of organelles and vacuoles. In these neuroblasts mitochondria are swollen and ruptured and remnants of the endoplasmic reticulum clump with remaining mitochondria into electron dense aggregates. Mitochondrial aggregates are seen in 25% of remaining fzy5032/fzy1 mutant neuroblasts. Elevated levels of reactive oxygen species and calcium ions are seen in fzy5032/fzy1 mutant neuroblasts compared to controls. Almost 40% of fzy5032/fzy1 mutant neuroblasts contain ubiquitin-conjugated protein aggregates, and these are virtually non-existent in controls.

fzyAC-10/fzy1 larval brains show progressive neuroblast loss.

Homozygous germline stem cell clones are lost more quickly from ovaries compared to control clones.

More than 80% of cells in the ventral epidermis (VE) are arrested at metaphase in late stage 11 mutant embryos (compared to less than 5% of VE cells in wild-type embryos) and 62% of the centriole pairs at the spindle poles are engaged. More than 95% of the cells in the VE are in mitosis in early and mid-stage 12 mutant embryos (there are virtually no cells in mitosis in the VE at these stages in wild-type embryos) and all centriole pairs are disengaged.

The maternal fzy contribution is stable enough to allow completion of mitotic proliferation in the dorsal epidermis, but not in the ventral epidermis. fzy1 embryos carrying the temperature-sensitive cdc2A171T in P{Dmcdc2A171T} at 18oC exhibit mitotic arrest at stage 11 in the ventral region, as expected. Temperature shift to the restrictive temperature causes a drastic reduction in the number of metaphase arrested cells.

Ventral epidermis absent, CNS degenerate, PNS absent.
