HDAC104556 heterozygote adults have increased lifespan and increased survival under paraquat-induced oxidative stress, as compared to wild-type controls.

Rpd304556 germline clones exhibit an abundance of γ-His2Av foci and evidence of a repair defect.

Rpd304556/+ flies have the normal number of dorsocentral bristles.

Similarly to wild-type controls, Rpd304556/+ heterozygotes show an age-dependent increase in susceptibility to temperature-sensitive paralysis.

Heterozygotes have no effect on the position effect variegation of w seen in the In(1)wm4h chromosome.

Heterozygous males show a 33% extension of life-span. Heterozygous females show a 52% extension of life-span. Calorific restriction does not result in a further extension of life-span in Rpd304556 heterozygotes.

Heterozygotes raised on medium containing trichostatin A (TSA) do not show an increase in the rate of lethality or developmental delay compared to wild-type flies raised on medium containing TSA and in fact the lethality caused by TSA is weakly suppressed in heterozygotes compared to wild type. Heterozygotes raised on medium containing trichostatin A (TSA) have a notched wing phenotype, with the notches increasing in number and size with increasing TSA concentration.

HDAC104556 homozygotes are lethal; HDAC104556/HDAC1303, HDAC104556/HDAC1313, HDAC104556/HDAC1326 and HDAC104556/HDAC1328 transheterozygotes are lethal under normal conditions, but the rare adults eclose and survive to short-lived and sterile adults in uncrowded yeast-supplemented conditions. When raised under carefully maintained culture conditions transheterozygotes of the genotype HDAC104556/HDAC1303 adult males eclose at 40% the expected rate and females at 30% the expected rate. Adults are short lived and females produce few eggs, apparently unfertilised. HDAC104556 has no dominant effect on PEV. Phenotypes include strong suppression of variegation in In(1)wm4, severely notched wings, small, malformed, sometimes curved or duplicated bristles, alulae that are larger than normal, and a reduction in the number of sex combs.

Embryos from mosaic females containing Rpd304556 homozygous germ-line clones, exhibit a pair rule phenotype in more than 80% of the majority (>65%) that fail to hatch. Partial or complete fusion of adjacent denticle belts are seen, resulting in embryos with 5 to 8 thoracic and abdominal segments. In addition variable, often severe, defects are seen in the head skeleton.

Embryos derived from Rpd304556 germline clones exhibit a variable pair-rule phenotype. They are also reduced in size.

Embryos derived from homozygous female germ line clones show a weak maternal effect, having pair rule segment defects.

Germline clones produce eggs with patterning defects: variable pair-rule segmentation defects.

Rpd304556/Snr1R3 double heterozygotes have wing vein defects; 32% have normal wings, 38% have an ectopic vein emanating from the posterior crossvein and 30% have ectopic veins emanating from the posterior crossvein and posterior to the L5 longitudinal vein. Rpd304556/brm2 double heterozygotes have a normal wing phenotype. The ectopic wing vein phenotype seen in Rpd304556/Snr1R3 and Rpd304556/Snr1E1 double heterozygotes at 29oC is partially suppressed by brm2/+.

Sir205327a does not show a dominant synthetic lethal interaction with Rpd304556.

The partial lethality due to runScer\UAS.cLa; Scer\GAL4nos.PG (3% viable) is partially suppressed by maternal heterozygosity for Rpd304556 (rescues to 31% viable).

The addition of Pc4 to Rpd304556 animals increases the average number of extra sex comb teeth per leg from 2.7 to 3.94. The addition of esc10 or E(z)63 to Rpd304556 animals has no effect on the extra sex comb phenotype.

The eye phenotype caused by expression of ttk69.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4hs.2sev is not dramatically modulated by Rpd304556.

Eggs from Rpd304556/groE48 mothers exhibit an embryonic lethality significantly higher than from either groE48/+ or Rpd304556/+ mothers. Cuticles from over 70% of unhatched embryos show a bicaudal phenotype, including a duplicated posterior spiracle, and a mirror image duplication of the posterior abdominal segments in place of the normal anterior segments. Eggs from Rpd304556/Df(3R)Espl22 mothers exhibit an embryonic lethality significantly higher than from either Df(3R)Espl22/+ or Rpd304556/+ mothers.
