Structures at the ventral midline and CNS development are defective. Posterior commissure is missing in every segment and anterior commissure formation in the thoracic segments is affected. Growth cones that pioneer the posterior commissure do not make the correct pathway choice and grow anteriorly and posteriorly after correct extension. Growth cones that pioneer the anterior commissure do make the correct pathway choice and grow straight toward the midline.
Embryos lack most protocerebral (P) neuroblasts and some deutocerebral (D) neuroblasts. Most of the anterior (Da) and posterior (Dp) deutocerebral neuroblasts, and some of the central (Dc) deutocerebral neuroblasts are missing. Most or all of the protocerebral Pc4, Pa3-6 and Pp cell groups are missing, while Pc1/2 and Pa1/2 neuroblasts are less or not at all affected. The protocerebrum is strongly reduced in size and there are some defects in the deutocerebrum in late embryos.
Many midline neurons are disrupted and mutants exhibit a reduction in the lateral CNS neurones in each abdominal hemineuromere.
