Detailed Description

Statement Reference Heterozygous rl[Sem] produces extra wing vein material. Absolute bouton number and bouton number per muscle area is significantly increased in heterozygous larvae compared to controls at the neuromuscular junction in segment A3 in muscle 6/7. Heterozygotes have ectopic veins in the wing. rlSem/+ flies have ectopic vein material in the wing. rlSem mutants exhibit extra wing veins. The wings of rl[Sem]/+ animals form scattered fragments of ecoptic vein tissue in intervein regions. Heterozygotes have a rough eye phenotype, which is enhanced if these flies also express rlGMR.T:Scer\GAL4,T:Hsim\VP16,T:Hsim\gD or rlGMR.T:SV40\nls2,T:Scer\GAL4,T:Hsim\VP16,T:Hsim\gD. rlSem flies have an extra wing vein phenotype. Additional R7 cells are formed in the eye. At stage 17 rlSem mutant embryos have an average of 6 midline glial cells per segment (n=231), compared to 2.8 per segment in wild-type. rlSem/+ mutants exhibit a significant increase in the number of synaptic boutons compared to wild-type. 10.7% of embryos derived from heterozygous females have less than 4 denticle belts. Extra wing veins develop. Heterozygous rlSem animals display a mild rough eye phenotype. Flies carrying rlSem have a rough eye phenotype. Heterozygous females show a weak dominant sterility; 10.2% of the eggs laid by these females fail to hatch and 20.0% of these nonviable embryos have defects similar to those associated with gain-of-function alleles of tor. Mutants display no apparent proliferation defect. Adult have moderately rough eyes and extra wing vein material. Shows supernumerary pigment cells in the developing eye, due to failure of apoptosis. rlSem/+ shows ectopic wing vein tissue. Ommatidia differentiate additional R7 cells, phenotype suppressed by P{sE-D-junS82A,T92A,T107A}. Rough eye phenotype is slightly suppressed by JrabZIP.hs.sev.T:Ivir\HA1. The sev signalling pathway is activated, even in the absence of the boss inducing signal: flies show positive phototaxis towards UV light. Eyes show multiple R7 cells. Embryos from heterozygous females show reduced numbers of abdominal segments though the head skeleton and filzkorper do develop. As for dominant Egfr mutants, heterozygotes have extra wing vein material. Clonal analysis reveals phenotypes in the adult including loss of wing vein, ectopic wing vein, reduced cell size, cell lethality and tergite bristle abnormalities. Rough eyes and extra wing veins. Formation of supernumerary R7 photoreceptor cells.

Genetic Interactions
| |||
Statement Reference S6kII[ign-Δ58-1]/Y ; rl[Sem]/+ double mutant larvae do not have a higher number of boutons at the neuromuscular junction than either single mutant. rl[Sem]/+ enhances the mutant wing phenotype caused by expression of Mad[Scer\UAS.cNa] under the control of Scer\GAL4[vg.int2.1]. The rlSem extra wing vein phenotype is unaffected by the expression of Mkp3Scer\UAS.cRa under the control of Scer\GAL4en-GAL4-33. The formation of additional R7 cells seen in rlSem flies is almost completely suppressed by the expression of Mkp3hs.sev. The increase in midline glial cells seen in rlSem mutant embryos is slightly enhanced by Wunspecified/Wunspecified (from 6 (n=231) to 6.6 (n=91)). The reduction in midline glial cell numbers seen in commΔe39 mutant embryos is suppressed in double mutants with rlSem. These rescued midline glial cells are scattered throughout the midline, which lacks axons in these animals. rlSem/+ alone is not sufficient to rescue the reduced eye and head phenotypes of flies that are homozygous for Ras85Dx7b in the eye disc. However, the addition of Ras85DD38E in combination with rlSem results in a substantial rescue of the Ras85Dx7b eye and head size phenotypes; differentiated photoreceptor cells are seen and a substantial fraction of the ommatidia contain the full complement of photoreceptors. The addition of one copy of SelDk11320 to rlSem/+ fliers has no effect on the eye phenotype. Suppresses the wing margin incision and loss of microchaetae phenotypes of lacek05305/lace2 flies. rlSem completely suppresses the PTP-ERhs.sev phenotype, to the point of producing ectopic R7 cells as seen in mutants in rlSem alone. At 25oC rlSem dominantly enhances most of the hyperplastic growth seen in Ras85DV12.Scer\UAS Scer\GAL4dpp.blk1 wing discs resulting in a much larger disc with severe disruptions of morphology. Ras85DV12.G37.Scer\UAS Scer\GAL4dpp.blk1 and rlSem act synergistically to produce a small amount of hyperplastic growth in third instar wing discs. Mutants die as pharate adults at 25oC and have deletions of tarsal segments and extra wing vein material along L3. No ectopic proliferation is seen with Ras85DV12.C40.Scer\UAS. The adults often have wing blisters and extra vein material along L3. This phenotype is not suppressed by phlXS-2541. TfIIA-SE73 suppresses the rough eye phenotype of sevS11.T:Hsap\MYC, phl::tor12D.hs.sev, Ras85DV12.sev, rlSem and svp2.sev. Enhancer of the dosage-dependent (two or more copies of P{sev-svp1} or P{sev-svp2}) transformation of cone cells into R7 photoreceptors and at a lower frequency R7 cells into outer photoreceptors. Eyes show multiple R7 cells, even in a sev- mutant background. rlSem suppresses the reduced viability associated with phl12, phl1 and phl107, and the eye phenotype of phl1. Homozygous tll1 embryos from rlSem/+ mothers have a tll mutant phenotype. The extra wing vein phenotype of heterozygotes is enhanced in transheterozygotes with dominant Egfr alleles. Flies heterozygous for EgfrE3 or EgfrE1 and rlSem have smaller eyes than EgfrE3 or EgfrE1 heterozygotes alone.

Statement Reference The ectopic wing vein phenotype seen when Hsap\ATN1N917.65Q.Scer\UAS is expressed under the control of Scer\GAL4GMR.PF is enhanced in rlSem heterozygotes.
