The mutated residue G175 is strictly conserved in, and specific for, the CKI kinase family. G175 falls in the L-9D loop. This lesion may weaken or prevent the interaction with at least some of the dco substrates, without affecting its catalytic activity.

The wings of dco2/dco5B2.6 flies show polarity defects in the region proximal to the anterior cross vein. The eyes of these transheterozygotes also show significant polarity defects.

dco3 in combination with dco2 or dco18 : lethality in larval or early pupal stages, with overgrown discs. dco2 in combination with dco18 : show lethality during larval or early pupal stages, discless phenotype. dco2 in combination with dcole88 : show lethality during larval or early pupal stages. dco2 in combination with dcoi3-193 or dcoS139602 : show lethality during larval or early pupal stages, with abnormal discs with granular zone and abnormal folding. dco2 in combination with dcoS053813 : show lethality during larval or early pupal stages with small discs. dco2 in combination with dcoS091510 : show lethality during early or late pupal stages, with abnormal discs with granular zone and abnormal folding. dco2 in combination with dcoP103 : adult viable with expanded wings and leg outgrowths. dco2 in combination with dcoS144701 : adult viable with normal phenotype.

When raised at 25oC the larval period of dco2 flies in heteroallelic combinations is greatly extended and they develop overgrown imaginal discs. Imaginal discs show gap-junctional communication defects: coupling is severely diminished. The single layer epithelium is maintained during overgrowth and they differentiate into the correct but abnormal adult structures. Embryos are smaller than wild type at 4-5 days after egg laying: ventral and dorsal arms and ventral plates of head skeleton are reduced.
