      Construct: w+mC minigene altered to include a su(Hw) binding region (all 12 binding sites, nucleotides 647 to 1077 of Marlor et al., Mol Cell Biol 6:1129-1134), upstream of the w eye enhancer (nucleotides -1084 to -1465 of Qian et al., Genetics 131:79-90), upstream of the w coding region, upstream of a second su(Hw) binding region.
      Two gypsy\su(Hw)BR binding sites flank the w+mET sequences to insulate the w gene from chromosomal position effects.
      P{SUPor-P-lt} contains a mini-w gene that has two flanking gypsy\su(Hw)BR sites, one of which is upstream of the w eye promoter.

      The eye colour is not affected if wBR.E.BR flies are also carrying e(y)11 and zv77h. Flies that are also carrying e(y)11, zv77h and su(Hw)2/su(Hw)V have reduced eye pigmentation.
      Expression of wBR.E.BR is reduced (in a background of reduced su(Hw)+) by the expression of tethered Pc, Scm or esc. In the case of Pchs.T:su(Hw)-ZnF,T:Zzzz\FLAG, the effect is seen without the use of heat shock. For Scmhs.T:su(Hw)-ZnF,T:Zzzz\FLAG and eschs.T:su(Hw)-ZnF,T:Zzzz\FLAG heat shock must be applied daily. In the case of eschs.T:su(Hw)-ZnF,T:Zzzz\FLAG the effect is dependent on the site of insertion of P{SUPor-P}. ph-p409 completely suppresses the strong silencing of wBR.E.BR in P{SUPor-P}22F caused by tethered Pc, and substantially suppresses the strong silencing of wBR.E.BR in P{SUPor-P}22F caused by tethered Scm or esc.

      Three insertion lines carrying this allele show variegation for w, and behave as though the insertions are heterochromatic. In situ hybridization confirms that the insertion in one of these lines is in the second chromosome centric heterochromatin. In a su(Hw) mutant background the variegating phenotype of these insertions becomes more pronounced, and the su(Hw) mutation acts dominantly in this interaction.
