Sterile females show rough eyes and clipped wings. Stage 14 oocytes of sub1794/subDub show a defect in spindle pole formation - most often spindles are either monopolar or tripolar, karyosome may be split, spindles broken or misshapen such that they fray, or do not taper at the poles. Oocytes of subDub/+ females show relatively minor defects in spindle organization.
High levels of nondisjunction occur in both divisions. Anaphases are uneven or unipolar and many figures show lagging chromosomes.
Extensive cell death. hemizygotes are semi-lethal.
Restrictive temperature is 25oC. In mutant females both non-exchange and exchange chromosomes undergo nondisjunction, but the effect on nonexchange chromosomes is more pronounced. Recombination levels are reduced slightly. Multiple nondisjoined chromosomes frequently cosegregate to the same pole. Nondisjunction of all chromosomes occurs in meiosis I of males, though levels are lower than in females. Homozygotes exhibit phenotypes consistent with cell death. Rare escapers at 25oC are very short lived and show rough eyes, etched tergites, crumpled or nicked wings and bristles either missing or duplicated. At 18oC homozygous progeny are more common, though at most 20% of the expected number of homozygotes eclose. Heterozygous mothers generate gynandromorph progeny. Heterozygous parents generate 8-12% embryonic lethality, probably due to autosomal aneuploidy of offspring. Homozygous mutant larvae are lethargic, rarely wandering or pupating outside the food. Some imaginal discs are missing nad remaining discs are reduced in size. Brains are normal in size. Larval neuroblast squashes reveal no defect in mitosis. Pupal lethal phenotype includes melanotic tumors, rough eyes, missing or duplicated bristles and missing body parts, suggesting random cell death.
Allelic series (strongest to weakest phenotype): sub131/subDub = sub202/subDub = sub218/subDub = sub22/subDub = sub72/subDub = sub104/subDub = sub158/subDub > sub1794/subDub > +/subDub.
Frequency of nondisjunction in abo1/subDub is double that of subDub/+, however no increase in nondisjunction is observed in abo2/sub females, suggesting that the interaction of subDub with abo1 is allele-specific or due to a locus elsewhere on the chromosome. subDub is likely to be either antimophic or neomorphic.
