Flies expressing wgl-12.Scer\UAS under the control of Scer\GAL4NP7011 (and shifted to the permissive temperature at the third instar larval stage) exhibit a segment gap phenotype in the abdomen.

wgl-12.Scer\UAS expression in all midline cells from late stage 9, driven by Scer\GAL4sim.PS, does not interfere with midline cell differentiation.

Considerable levels of ectopic apoptosis of photoreceptors occurs throughout the developing eye at 42 hours after puparium formation (APF) in wgl-12.Scer\UAS; Scer\GAL4GMR.PF pupae raised at 25oC and switched to 16.5oC (the permissive temperature) at 24 hours APF.

Expression of wgl-12.Scer\UAS under the control of Scer\GAL430A rarely has any effect on axillary sclerite 3 in the wing hinge.

Expression of wgl-12.Scer\UAS under the control of Scer\GAL4byn-Gal4 results in an enlargement of the lumen of the hindgut (except for a short anterior portion corresponding to the small intestine), and there is no morphological boundary between the large intestine and rectum.

Expression of wgScer\UAS.cLa under the control of Scer\GAL4da.G32 at 22oC results in expansion of 4o cells at the expense of 3o cells in the dorsal epidermis of the embryo (in wild-type embryos each parasegment contains 4 types of cells in the dorsal epidermis; the 1o, 2o and 3o cells in the anterior half of the parasegment and the 4o cells in the posterior half of the parasegment). Expression of wgScer\UAS.cLa under the control of Scer\GAL4da.G32 at 18oC (a more permissive temperature for the mutant wg protein) produces a similar effect. Expression of wgScer\UAS.cLa at 6 hours of development results in a further loss of the anterior half of the parasegment, while later expression produces little effect.

When wgl-12.Scer\UAS is driven by Scer\GAL4Dll-981 at 16oC, most of the animals die and only a few escapers are observed. When reared at 25oC defects are seen in distal structures. In the antenna all three sensilla types can be seen, but there is a significant increase in the coeloconic sensilla numbers, which appear in clusters over the antennal surface. When third instar larvae are shifted to 16oC the antennal disc morphology is not significantly compromised.

Expression of wgl-12.Scer\UAS driven by Scer\GAL4sd-SG29.1 at 17oC has little effect on the development and patterning of the wing in the third instar larva. Adult wings are wild-type in size but crumpled with a few blisters, some extra bristles and enlarged veins.

Expression driven by Scer\GAL4arm.PS beginning (by shift down in temperature) at mid stage 11 (6.5hr AEL) causes most cells to secrete naked cuticle. Expression driven by Scer\GAL4arm.PS beginning (by shift down in temperature) at late stage 11 causes cells in the midventral (rho expressing) domain to secrete naked cuticle.

Ommatidial differentiation is impaired in eye discs expressing wgl-12.Scer\UAS under the control of Scer\GAL4dpp.blk1.

Embryos expressing wgl-12.Scer\UAS under the control of Scer\GAL4arm.PS do not form ventral denticles.

Overexpression of wgl-12.Scer\UAS under the control of Scer\GAL4Bx-MS1096 results in ectopic wing vein formation and the formation of margin bristles in the wing blade.

Expression of wgl-12.Scer\UAS under the control of Scer\GAL4prd.RG1 in sgl08310 embryos does not generate naked cuticle.

Scer\GAL4sd-SG29.1-mediated expression produces flies bearing wings with a high degree of blistering, thickened veins and extra bristles. The distribution of trichomes in the wing blades reveals a very uneven cell density.

Continual Scer\GAL4dpp.blk1 induced expression during the larval period at 25oC causes no pattern abnormalities or transdeterminations in the adult. Raising larvae at 25oC until 50 hours AEL then shifting to 15oC results in transdetermination in the adult; maxillary palp to antenna, antenna to maxillary palp and leg to wing. The eyes are smaller due to loss dorsally and ventrally. Continual Scer\GAL4ptc-559.1 induced expression during the larval period at 25oC causes no pattern abnormalities or transdeterminations in the adult. Raising larvae at 25oC until 50 hours AEL then shifting to 15oC results in transdetermination in the adult; maxillary palp to antenna, antenna to maxillary palp, labial to antenna, head to wing, leg to wing and genital to maxillary palp.

Scer\GAL4h-1J3 induced expression partially restores naked cuticle in alternate segments of smo- embryos.

Loss of slou expression normally seen in wgl-17 mutants is rescued by Scer\GAL4 line 69B acting on P{UAS(-FRT)wg.ts}, slou-expressing cells are in the appropriately positioned clusters and the epidermis phenotype is substantially rescued. wg expression in the mesoderm of wgl-17 mutants, driven by P{GAL4-twi.B} expression, is capable of restoring some ectodermal en expression as well as partially rescuing the cuticle mutant phenotype.

Ectopic expression at the posterior margin of the eye disc delays normal furrow initiation. The central region of the eye disc is less affected so that initiation occurs in the centre ahead of more lateral regions. The eye eventually develops almost normally so the block imposed by wg is not permanent.

In combination with Scer\GAL4h-1J3, at 16oC, embryos show naked cuticle in place of alternate denticle belts. When shifted to 16oC from 25oC during first larval instar the bristles that mark the ventral lateral surface are expanded, elements of the dorsal surface are missing, the leg circumference is increased and the claw is often duplicated. Ventralmost pattern elements are not expanded. Phenotypes of legs that arise from discs exposed to ectopic wg beginning at first, second or third instar are comparable. BrdU experiments on leg discs reveal that dorsal cells over-proliferate, though largely outside the region of ectopic wg expression. In combination with Scer\GAL4dpp.blk1, leg phenotypes include loss of dorsal structures, increase in circumference and fusion of leg segments. The claw is sometimes duplicated. The proximo-distal axis is intact. Bifurcations sometimes occur.

When driven by Scer\GAL4h-1J3, the overall size and diversity of denticles are partially restored in pair rule "mirrors" to wgen11 mutants. However naked cuticle is not recovered. When driven by Scer\GAL4prd.RG1, aspects of all the segmental pattern defects of wgen11 are recovered: size, denticle diversity and naked cuticle. These cuticles have four denticle bands, each much wider than the wild type bands, but each containing all denticle types, in roughly correct order.

Expression of wgl-12.Scer\UAS in animals expressing Cul-5EP3390 in the wing and haltere disc under the control of Scer\GAL4sd-SG29.1 partially rescues the wing-blade defects observed at 29[o]C.

wgl-12.Scer\UAS; Scer\GAL4dpp.blk1 suppresses duplication of the wing pouch and prospective notum in ds38k homozygous wing discs, although the hinge domain is expanded compared with the wild type. Normal anterior-posterior subdivision is also restored in these discs. These animals die before reaching adulthood.

Co-expression of wgl-12.Scer\UAS can rescue both the wing notching phenotype and loss of sensory bristles caused by expression of dlpScer\UAS.cBa under the control of Scer\GAL4C96. In addition, ectopic margin bristles close to the wing margin are seen in these flies.

Coexpression of wgl-12.Scer\UAS and fz2Scer\UAS.cCa under the control of Scer\GAL430A results in a diminution of axillary sclerite 3 in the wing hinge.

The addition of wgl-12.Scer\UAS suppresses the wing pouch phenotype seen in NECN.Scer\UAS/Scer\GAL4sd-SG29.1 wing discs, and abolishes the cell death phenotype. Wings in these adult flies are almost wild-type in size and in many instances display a partial restoration of bristles at the edge. In addition these wings display enlarged veins which are much broader than those produced by either NECN.Scer\UAS or wgl-12.Scer\UAS alone. The addition of wgl-12.Scer\UAS strongly suppresses the wing phenotype seen in NECNΔ10-12.Scer\UAS/Scer\GAL4sd-SG29.1 flies, restoring the wing almost to wild-type, and partially suppresses the wing phenotype seen in NECNΔ17-19;24-26.Scer\UAS/Scer\GAL4sd-SG29.1 flies. The addition of wgl-12.Scer\UAS (driven by Scer\GAL4sd-SG29.1) to Df(3R)Dl-FX3/+ flies leads to a dramatic increase in the width of the wing veins.

The lack of denticles seen in embryos expressing wgl-12.Scer\UAS under the control of Scer\GAL4arm.PS is suppressed by injection of both dsRNA produced by annealing fz374-2360 and fza.374-2360 RNA and dsRNA produced by annealing fz2L.cKa and fz2a.L.cKa RNA; a pattern of denticles similar to wild-type is restored and ectopic denticles are produced in regions where naked cuticle should be secreted.

Scer\GAL4dpp.blk1-mediated expression in vg83b27-R mutants fails to rescue loss of wing tissue.

sgl08310 embryos expressing wgl-12.Scer\UAS under the control of Scer\GAL4prd.RG1 show a minor rescue of the sgl08310 cuticle phenotype, although naked cuticle is not specified.

Coexpression of wgl-12.Scer\UAS and SerScer\UAS.cSa with Scer\GAL4sd-SG29.1 the resulting flies bear large wings that are covered in bristles. The wings lack a defined margin and are more round that elongated along the PD axis. Coexpression with Scer\GAL4ptc-559.1 leads to the production of extra wings off the proximal dorsal side of the wing.

Scer\GAL4ptc-559.1-mediated coexpression with SerScer\UAS.cSa causes outgrowth on the ventral side of the wing discs and a small amount on the dorsal side but the nick in the wing margin is repressed. Ectopic wing tissue is also found in the compound eye and legs. Scer\GAL4h-540.3-mediated expression in SerRX82/Df(3R)D605 hemizygous second/early third larval instar fails to rescue the rudimentary wing phenotype.

Scer\GAL4arm.PS mediated expression of wgl-12.Scer\UAS in wgl-17 Df(2R)enE embryos at varying temperatures can be used to study the dose response to wg for A cells. At 17.5oC wgl-12.Scer\UAS produces the same phenotype as when wild type wg (wgScer\UAS.cLa) is added. At 20oC there are only a few ventral denticles and some beard in T1. At 22oC there are two lateral stripes of ventral denticles and some beard in T1. At 23oC the abdomen is completely covered in denticles but there is no beard in T1. At 25oC the abdomen is completely covered in denticles, weak thoracic denticles are present, but there is no beard in T1. At 28.5oC embryos are indistinguishable from wgl-17 Df(2R)enE embryo.

Scer\GAL4ptc-559.1 induced co-expression of wgl-12.Scer\UAS and SerScer\UAS.cSa causes a duplicated pair of extra wings that arise from the proximal dorsal side of the endogenous wings. The endogenous wings may exhibit extra margin material. Wing tissues appear, with characteristic marginal bristles, from the proximal dorsal side in all legs. Scer\GAL4h-1J3 induced coexpression of wgl-12.Scer\UAS and SerScer\UAS.cSa fails to cause ectopic wings, expression of NScer\UAS.cBb with both wgl-12.Scer\UAS and SerScer\UAS.cSa causes ectopic wing development.
