P4c6 Poxn cDNA expression is driven by an XbaI-KpnI fragment of the first 95bp of Hsp70 leader and 250bp of Hsp70 upstream sequence and a 1.2kb sev enhancer fragment. Transcription is terminated by a 0.8kb tubulin fragment.

When Poxnhs.sev is heatshock induced for 30 minutes at 37oC at 8 hours APF, mechanosensory bristles are transformed into chemosensory bristles.

Flies expressing Poxnhs.sev have a rough eye phenotype. The arrangement, shape and number of cone cells in mid-pupal eye discs is disturbed.

Overexpression causes development of ectopic chemosensory organs in the larva and adult.

Rough eye phenotype unaffected by TfIIA-SE73.

Expression during embryogenesis leads to the formation of an excess of p-es organs (FBrf0055546) due to transformation of many ch and m-es organs into p-es organs.

Induction 10 hours after puparium formation (APF), at the time when the mechanosensory SMCs emerge, causes morphological transformation of mechanosensory bristles into chemosensory bristles. Ultrastructure analysis of the shaft reveals it comprises of two lumens: a tubular compartment with contains two to five dendrites and a crescent-like compartment which contains no dendrites. Inducing expression 14 and 15 hours APF results in transformation of the external morphology of some mechanosensory bristles. No transformation is observed when expression is induced 16 hours APF or later. DiI labelling of sensory neurons at 16 and 20 hours APF shows a few cases of mechanosensory bristles innervated by several neurons and more frequently a group of unconnected neurons that do not connect to the external shaft.

Embryos display supernumerary kolbchen in all thoracic segments.

When animals containing Poxnhs.sev and phlF20.Scer\UAS (driven by Scer\GAL4hs.PB) and heatshocked, an additive phenotype is seen, mechanosensory bristles are transformed into chemosensory bristles and all epidermal cells are transformed into bracts. A similar effect is seen by the combination of Poxnhs.sev and Poxnhs.sev (driven by Scer\GAL4hs.PB).

The svspa-pol eye phenotype is enhanced by Poxnhs.sev, with the eye lacking all lenses and most bristles. The number of cone cells present in mid-pupal eye discs is reduced compared to svspa-pol single mutants and those that are seen are smaller than normal and seem to be undergoing apoptosis. One copy of svsev.PJ or sv+mFa rescues the Poxnhs.sev eye phenotype to wild type.
