Statement Reference When mutant third instar larvae are subjected to heat treatment, their wing and leg imaginal discs show a phenotype in the morphology of the golgi area. The proportion of membrane that forms cisternae increases from about 30% (in heat shocked controls) to about 56%. 60% of these are stacked compared top about 30% in controls. comt1 and comt4 flies bearing P{hsp70-NSF} exposed to heat shock, followed by one day recovery, are highly resistant to exposure to 38oC while their comt- siblings lacking the transgene remain sensitive. | |||

| Comments | Expression of comt[hs.PP] using heat shock can rescue the larval lethality of comt[clp]. Maximal rescue of the larval lethality is obtained if comt[hs.PP] is expressed between 60 and 72 hours after egg laying. Although rescue results in mature pupae, none of the rescued animals eclose as normal flies.
Expression of comt[hs.PP] in comt[clp] flies throughout larval development (to rescue the larval lethality of comt[clp]), followed by a second heat shock after puparium formation results in normal adults, provided that comt[hs.PP] is activated 4 days prior to eclosion (activation prior to this period results in progressively reduced rescue, depending on the time of induction).
The reduction in the number of boutons at the larval neuromuscular junction that is seen in comt[clp] mutants is rescued by expression of comt[hs.PP]. | ||
