ClaI genomic fragment containing 142bp of the Sry-β-janA intergenic region is fused to Ecol\lacZHsp70Bb.HZ50. Fragment corresponds to Sry-β sequences from -140bp to +2bp (where +1 refers to the Sry-β transcription start site).
A EcollacZ construct driven by -140 to +2 of Sry-β (+1 is the Sry-β transcription start site) drives testis-specific expression of EcollacZ. In both the third instar larva and the adult male, expression is not seen in the apical cells of the gonad. In the adult male, faint expression is detected in premiotic spermatocytes, and high levels are detected post-meiotically.
