Somatic clones of rhoScer\UAS.cGa; Scer\GAL4Act5C.PP cells in the follicle cells produce either ectopic dorsal appendages or ectopic chorionic material resembling warts. These warts are restricted to the the ventral/lateral region in the middle of the egg chamber, whereas ectopic dorsal appendages only form in the ventral/lateral region around the anterior circumference of the egg chamber (the collar region). These ectopic appendages include many cells recruited by the clone - clones of just 2 rhoScer\UAS.cGa; Scer\GAL4Act5C.PP cells can induce ectopic appenadge formation.

rhoScer\UAS.cGa; Scer\GAL4elav.PLu embryos have increased numbers of midline glial cells.

Clones in the prospective notum or wing hinge in the wing disc expressing rhoScer\UAS.cGa under the control of Scer\GAL4αTub84B.PZ have wiggly borders.

Clones in the wing disc (generated during the first larval instar) expressing rhoScer\UAS.cGa under the control of Scer\GAL4αTub84B.PZ can result in dramatic reorganisation of the wing disc, including the formation of ectopic dorsal compartments and ectopic notum primordia or the abnormal expansion of the dorsal compartment.

Ectopic expression of rhoScer\UAS.cGa under the control of Scer\GAL4E4 in a wild-type background has no visible effect on chorion structure.

Eggs derived from females expressing rhoScer\UAS.cGa under the control of Scer\GAL4T155 are dorsalised.

Leg discs in which rhoScer\UAS.cGa is expressed under the control of Scer\GAL4sca-109-68 have an increased number of sensory organ precursor cells in the chordotonal cluster. The proneural cluster in the ectoderm is reduced or absent and the invaginating stalk of cells is much thicker than wild type.

Scer\GAL455B-mediated expression causes the production of dorsal appendages around the anterior circumference of the egg. A small fraction of the eggs are fertilised and develop into dorsalised embryos (absence or complete elimination of denticle bands and expansion of dorsal hairs).

Embryos expressing rhoScer\UAS.cGa under the control of Scer\GAL4en-e16E have ectopic denticles anterior to the denticle belts.

Somatic clones of Scer\GAL4Act5C.PP cells in the eye disc in animals carrying rhoScer\UAS.cGa and spis.Scer\UAS or spiScer\UAS.s.T:Avic\GFP cause ectopic photoreceptor differentiation. This phenotype is completely suppressed by raspT392/raspT802.

Expression of rhoScer\UAS.cGa, under the control of Scer\GAL4ptc-559.1, in SerRX82 embryos raised at 18 or 22oC rarely restores proper denticle row 4 polarity. However, raising embryos of the same genotype at 29oC does restore row 4 polarity and causes an expansion of type 5 denticles posterior to the normal denticle field.

Clones expressing rhoScer\UAS.cGa under the control of Scer\GAL4αTub84B.PZ induced in the first larval instar can stimulate proliferation in vnL6/vnddd-4 wing discs, irrespective of the position of the clone within the wing disc.

When rhoScer\UAS.cBa and SScer\UAS.cBa are driven by Scer\GAL4hs.PB and heatshocked, extra bracts are seen to associate with a few of the mechanosensory bristles, but not chemosensory bristles.

The ventralised phenotype of eggs derived from females expressing kek1Scer\UAS.cGa under the control of Scer\GAL4T155 is not altered if the females are also coexpressing rhoScer\UAS.cGa.

Leg discs in which both rhoScer\UAS.cGa and Dl::NΔECN.Scer\UAS are expressed under the control of Scer\GAL4sca-109-68 have a phenotype resembling that of rhoScer\UAS.cGa expressed under the control of Scer\GAL4sca-109-68 rather than that of Dl::NΔECN.Scer\UAS expressed under the control of Scer\GAL4sca-109-68.

SIIN mutation dramatically suppresses the dorsal appendage phenotype, appendages appear more normal and the embryos displays reduced but distinct ventral denticle belts. spi mutation has no effect on the dorsal appendage phenotype. Expression in grk mutants reduced the severity of the phenotype, some ectopic dorsal appendage material is produced and embryos form denticle belts.

Comments

rhoScer\UAS.cGa driven by Scer\GAL4rho.654 rescues the oenocyte defect but not the secondary sensory organ precursor cell defects associated with rho7M43.
