Description

UASt regulatory sequences drive expression of a dominant negative form of shg (consists of the C-terminal 208 amino acids, which encode the transmembrane and cytoplasmic domains). The protein is tagged at the N-terminal end with the Tag:SS(aos) signal sequence and is also tagged with Tag:MYC.

The expression of shgi.Scer\UAS.T:SS-aos,T:Hsap\MYC under the control of Scer\GAL4dpp.PU in third instar larval wing disc cells results in some expressing cells migrating and crossing the anterior-posterior compartment border; the clonal expression of shgi.Scer\UAS.T:SS-aos,T:Hsap\MYC under the control of Scer\GAL4Act5C.PI leads to expressing cells migrating and scattering across long distances in the wing disc, and frequently exhibiting long actin rich protrusions, as compared to controls.

Expression of aos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en.PU results in loss of wing blade.

When argos::shgi.Scer\UAS.T:Hsap\MYC is driven by Scer\GAL4pnr-MD237, abnormal patterning of the notal microchaetae is seen. Instead of being aligned in five straight rows, as seen in wild-type, they are arranged chaotically. The density of bristles is also increased. When argos::shgi.Scer\UAS.T:Hsap\MYC is driven by Scer\GAL4neur-GAL4-A101 or Scer\GAL4sca-537.4 no effect is seen on bristle pattern.

Overexpression of argos::shgi.Scer\UAS.T:Hsap\MYC in all cells of the medial half of the notum, results in a slight decrease in bristle density.

When argos::shgi.Scer\UAS.T:Hsap\MYC is driven by Scer\GAL4en-e16E wing margin defects are seen in the adult wing.

Expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E results in posterior wing notching, mimicking wg hypomorphic mutants. The strength of the wing notching phenotype caused by expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E varies depending on the particular P{UAS-shg.CADi} insertion used. Flies carrying the P{UAS-shg.CADi}10 insertion expressed under the control of Scer\GAL4en-e16E always retain the posterior crossvein and a tuft of marginal bristles between the tips of wing veins 4 and 5, while these structures are consistently absent in flies carrying the P{UAS-shg.CADi}5 insertion expressed under the control of Scer\GAL4en-e16E. Expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4GMR.PF results in rough eyes. Extra interommatidial bristles are produced and two bristles can often be seen originating from a single socket.

Scer\GAL4arm.T:Hsim\VP16 mediated expression causes head and dorsal defects. Scer\GAL4en-e16E mediated expression causes loss of wing margin in the posterior compartment. Scer\GAL4arm.PS mediated expression causes defects in adult appendages; notum duplications or a drastic reduction of the wing to a tiny winglet.

The loss of wing blade caused by expression of aos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en.PU is partially suppressed by Traf43/+.

The severity of the serrated wing margin phenotype in argos::shgi.Scer\UAS.T:Hsap\MYC; Scer\GAL4vg.PM flies is enhanced by twsj11C8/+.

The metastatic behaviour seen in clones in the eye disc which are homozygous for scrib1 and are also expressing Ras85DV12.Scer\UAS under the control of Scer\GAL4Act5C.PI is not suppressed if they are co-expressing argos::shgi.Scer\UAS.T:Hsap\MYC.

Overexpression of argos::shgi.Scer\UAS.T:Hsap\MYC in all cells of the medial half of the notum, results in a slight increase in bristle density in a scaBP2 mutant background.

The addition of pont5.1 enhances the wing margin phenotype seen in argos::shgi.Scer\UAS.T:Hsap\MYC, Scer\GAL4en-e16E flies. The addition of arm4 strongly enhances the wing margin phenotype seen in argos::shgi.Scer\UAS.T:Hsap\MYC, Scer\GAL4en-e16E flies. The addition of rept35 suppresses the wing margin phenotype seen in argos::shgi.Scer\UAS.T:Hsap\MYC, Scer\GAL4en-e16E flies.

The wing notching phenotype caused by expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E is enhanced by wgl-17, arm1, arm2 or dshb117. The wing notching phenotype caused by expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E is suppressed by sggM11. The rough eye phenotype caused by expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4GMR.PF is enhanced by wgl-17 and dshb117 and suppressed by sggM11. The wing notching phenotype caused by expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E is rescued by co-expression of armΔα.Scer\UAS though not by co-expression of Hsap\JUPScer\UAS.cWa.

Ectopic expression of armScer\UAS.cSa rescues the wing defect caused by expression of argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E to wild type. Loss of one copy of arm (arm1) enhances the wing phenotype of surviving flies expressing argos::shgi.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4en-e16E, most die as pupae.
