A constitutively active form of Mmus\Pkaca is expressed under the control of Scer\UAS regulatory sequences.
Scer\FRT-y+t7.7 cassette has been excised, juxtaposing Scer\GAL4 Scer\UAS binding sites with mutated Mmus\Pkaca coding sequences (mC*).

Expression of Mmus\Pkaca[mC.Scer\UAS] under the control of Scer\GAL4[ap-md544] is lethal.
Flies expressing Mmus\Pkaca[mC.Scer\UAS] under the control of Scer\GAL4[104Y] show reduced night-time sleep compared to controls.
Flies expressing Mmus\Pkaca[mC.Scer\UAS] under the control of Scer\GAL4[Mef2.247] show altered temperature preference behaviour compared to wild type: the avoidance index for high temperature is significantly reduced compared to wild type.
Mated females expressing Mmus\Pkaca[mC.Scer\UAS] under the control of Scer\GAL4[ppk.PG] show higher levels of sexual receptivity than control mated females. The rate of egg-laying of the mutant mated females is not significantly different from that of control mated females.
Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4elav.Switch.PO (induced using RU486) decreases daily sleep. Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4Tab2-201Y results in an increase in total daily sleep compared to controls. The adults flies have normal waking activity. Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4c309 results in an decrease in total daily sleep compared to controls. Activity normalised to waking time is not significantly higher than controls. Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL430Y or Scer\GAL4238Y promotes sleep during the day, but inhibits sleep during the night. Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4Mef2.247.Switch (in the presence of RU486) results in a reduction in sleep, due to a shortened sleep bout duration without a concomitant increase in the sleep bout number. When RU486 is withdrawn after about three days of sleep deprivation, these flies show a robust sleep rebound response.
Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4c309 results in reduced sleep compared to controls.
Postsynaptic expression of Mmus\PkacamC.Scer\UAS, under the control of Scer\GAL4eve.RN2, results in the averaged amplitude of synaptic currents recorded from whole-cell voltage clamp recordings of aCC/RP2 motoneurons being significantly increased compared with controls.
Mmus\PkacamC.Scer\UAS; Scer\GAL4en-e16E adults have blistered wings. There is precocious cell of wing blade cells in late stage pharate adults (G+; <3hrs before eclosion) of this genotype .
The ability of aCC/RP2 to fire action potentials (APs) in response to an injection of constant current is decreased when Mmus\PkacamC.Scer\UAS is constitutively expressed under the control of Scer\GAL4eve.RN2O and there is also a decrease in peak current density of voltage-dependent inward Na+ current compared to wild type.
7.3% of egg chambers have at least one binucleate nurse cell in females expressing Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4nos.UTR.T:Hsim\VP16. 3.1% of egg chambers have at least one binucleate nurse cell in females expressing Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4nos.PG.
When expression is driven by Scer\GAL4Itp-r83A-1664GAL4 in an otherwise wild type background larval molting is delayed. Molting delay is most evident at the L2 to L3 transition. Feeding Mmus\PkacamC.Scer\UAS,Scer\GAL4Itp-r83A-1664GAL4 larvae 20-hydroxyecdysone has no effect on the molting delay.
When expression is driven by Scer\GAL4ninaE.PD photoreceptors have slower macroscopic kinetics, similar to the effect caused by cAMP and IBMX.
Expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL471B results in a narrowing of the wing disc in the wing blade region.
Overexpression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4Mhc.PW significantly decreases quantal size as estimated by the average spontaneous miniature excitatory junctional current or miniature excitatory junctional potential recorded in muscle 6 in the absence of nerve stimulation in larvae. The number of boutons at the neuromuscular junction of muscles 6 and 7 is normal in larvae expressing Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4Mhc.PW.
Embryos expressing Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4e22c give rise to larvae with abnormal denticle belts; the wide posterior denticle rows of each segment are replaced by narrower and often incomplete rows of denticles with different morphology and polarity from wild-type.

Expression of Mmus\Pkaca[mC.Scer\UAS] under the control of Scer\GAL4[Oamb.4.4], using Scer\GAL80[ts.αTub84B] to limit expression to 3 days prior to mating, fails to suppress the ovulation phenotype seen in Oamb[unspecified] homozygotes.
The rough eye phenotype caused by expression of Hsap\MAPTV337M.Scer\UAS under the control of Scer\GAL4GMR.PF is enhanced by co-expression of Mmus\PkacamC.Scer\UAS; the size of the eye is reduced and necrotic foci are formed.
30% of egg chambers have at least one binucleate nurse cell in Akap200k07118a/Df(2L)N22-14 females expressing Mmus\PkacamC.Scer\UAS under the control of Scer\GAL4nos.UTR.T:Hsim\VP16.
The wing disc phenotype caused by expression of Mmus\PkacamC.Scer\UAS under the control of Scer\GAL471B is still seen in animals also co-expressing either ciWT.P.Scer\UAS.T:Ivir\HA1 or ci3m.Scer\UAS.T:Ivir\HA1. In contrast, in wing discs co-expressing ci5m.Scer\UAS.T:Ivir\HA1 and Mmus\PkacamC.Scer\UAS under the control of Scer\GAL471B, overgrowth of the wing blade region is seen, as in animals expressing ci5m.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL471B.
When expressed with Pka-R1BDK.Scer\UAS or Pka-R1BGD.Scer\UAS under the influence of Scer\GAL4E62-2, Scer\GAL4h-1J3 or Scer\GAL4C166-2, Mmus\PkacamC.Scer\UAS completely suppresses adult pattern defects elicited by Pka-R1BDK.Scer\UAS or Pka-R1BGD.Scer\UAS. Adults have normal legs, blistered wings, no cuticular outgrowths or duplicated wings.
