Expression of DlScer\UAS.cJa under the control of Scer\GAL448Y results in a 20% increase in the number of hub cells in the gonad in male larvae compared to controls. Expression of DlScer\UAS.cJa under the control of Scer\GAL4twi.PG results in a 14% increase in the number of hub cells in the gonad in male larvae compared to controls.
Scer\GAL4GMR.PF-driven expression of DlScer\UAS.cJa leads to an adult eye that is glossy and reduced in size with irregular ommatidial spacing. Third instar eye discs have excess R7 cells per ommatidium, while the average number of cone cells per ommatidium is significantly reduced at 24 hr after puparium formation.
Expression of DlScer\UAS.cJa under the control of Scer\GAL4elav-C155 causes no obvious defects in the pattern of the major axon tracts in the embryo.
Clones of cells expressing DlScer\UAS.cJa in the eye disc show substantial overgrowth compared to surrounding wild type tissue. Extra BrdU incorporation (a marker of S phase) and additional mitotic figures are associated with these clones, and a significant number of mitotic cells further posterior to the morphogenetic furrow (MF) are also observed. The MF is accelerated in these eye discs. Expression of DlScer\UAS.cJa under the control of Scer\GAL4GMR.PF produces no significant increase of proliferation in the eye disc compared with controls.
DlScer\UAS.cJa; Scer\GAL4fkh.14-3 causes ectopic cell movements in the developing proventriculus.
Expression of DlScer\UAS.cJa under the control of Scer\GAL4byn-Gal4 in embryos strongly suppresses boundary cell differentiation at the normal position in the hindgut. Occasionally, a few clusters of ectopic boundary cells are seen.
Clones in the eye disc expressing DlScer\UAS.cJa under the control of Scer\GAL4Act5C.PI can induce neural differentiation in a limited band of cells near the morphogenetic furrow.
Somatic clones of DlScer\UAS.cJa expressing cells (driven by Scer\GAL4Ubx.PdC) in the wing pouch of the dorsal surface produce outgrowths in the adult wing. Somatic clones of DlScer\UAS.cJa expressing cells (driven by Scer\GAL4Act5C.PI) in the wing pouch of both surfaces produce outgrowths in the adult wing. Dorsal outgrowths are associated with the differentiation of the wing margin structures, the outgrowths induced in the ventral compartment almost never differentiate wing margin elements. The rare cases where margin structures differentiate ventrally were all clones close to the endogenous margin. Large outgrowths also appear in the proximal regions in the legs and in the notum. In these regions, the clones differentiate ectopic sensory elements characteristic of the position where they appear. Somatic clones of DlScer\UAS.cJa expressing cells (driven by Scer\GAL4unspecified) in the wing disc produce clones that are in general very large. However clones close to the dorsal-ventral boundary are smaller than clones in proximal regions of the wing blade or close to the anterior posterior boundary. The size of the eventual outgrowth depends on the distance between the place where the clones are generated and the tip of the wing, rather than on the size of the clone. The numbers of cells in mitosis (metaphases and anaphases) is higher in DlScer\UAS.cJa ectopic expressing regions than in controls.
Clones of cells in the wing expressing DlScer\UAS.cJa under the control of Scer\GAL4Act5C.PI 0-6 hours after puparium formation result in the prevention of differentiation of some wing veins (L2, L4 and L5).
Expression of DlScer\UAS.cJa under the control of Scer\GAL4bs-1348 leads to a loss of wing veins. Expression of DlScer\UAS.cJa under the control of Scer\GAL4dpp.blk1 results in small, misshapen wings and ectopic wing margins.
In a wildtype background the expression of DlScer\UAS.cJa driven by Scer\GAL4twi.PG appears to have no effect on the embryo - there is no effect in CNS cell fate specification.
Flies expressing DlScer\UAS.cJa under the control of Scer\GAL4l(3)31-1-31-1 show defects in bristle development; the appearance of a second socket-like structure at the expense of the bristle shaft is seen in many bristles. This phenotype is seen in notal macrochaetae, wing hinge and wing margin bristles, leg bristles (including sex combs), interommatidial bristles, abdominal tergite bristles and other sensory bristles. The transformation does not affect all notal macrochaetae equally, ranging from 99% of anterior postalar macrochaetae being affected to no alteration of anterior dorsocentral macrochaetae. Few effects on notal microchaete development are seen.
Scer\GAL4ptc-559.1-mediated expression causes cell proliferation in both dorsal and ventral compartments of the wing imaginal disc displacing the dorsal ventral margin. Scer\GAL4Ser.PGF-mediated expression causes overproliferation of cells in the dorsal compartment of the wing imaginal disc.
Expression via Scer\GAL4Ser.PGF results in dorsal outgrowths of the wing imaginal disc and adult wings with excess dorsal wing blade material.
When expression is driven by Scer\GAL4bs-1348 wing vein loss is observed, with variable reductions in wing veins L2, L4 and L5.

Co-expression of AmunScer\UAS.cSa results in suppression of the adult rough eye phenotype and the reduced number of cone cells per ommatidium seen in Scer\GAL4GMR.PF, DlScer\UAS.cJa flies.
Scer\GAL4GMR.PF-driven co-expression of Su(H)Scer\UAS.T:Hsim\VP16 and DlScer\UAS.cJa results in an expansion of the zone of BrdU incorporation (a marker of S phases) and an increase in the number of cell divisions. Co-expression of Su(H)Scer\UAS.T:Hsim\VP16 and DlScer\UAS.cJa gives the same degree of excess proliferation in the eye disc as expression of Su(H)Scer\UAS.T:Hsim\VP16 alone. Co-expression of DlScer\UAS.cJa results in a moderate but reproducible and statistically significant increase in the number of mitoses seen in eye discs expressing E2fScer\UAS.cNa and DpScer\UAS.cDa under the control of Scer\GAL4GMR.PF. Co-expression of DlScer\UAS.cJa does not significantly modify the excess mitoses phenotype caused by Scer\GAL4GMR.PF-driven expression of CycEScer\UAS.cLa or CycAScer\UAS.cWa.
Clones in the eye disc expressing both dppScer\UAS.cSa and DlScer\UAS.cJa under the control of Scer\GAL4Act5C.PI result in neural differentiation in all regions anterior to the morphogenetic furrow. Neural differentiation is induced in all the cells surrounding the clone. Clones in the eye disc expressing both brkScer\UAS.cLa and DlScer\UAS.cJa under the control of Scer\GAL4Act5C.PI prevent furrow initialisation when they are at the posterior margin of the disc, as occurs in clones expressing brkScer\UAS.cLa alone under the control of Scer\GAL4Act5C.PI.

The transformation of the trichogen to a tormogen fate in the notal macrochaetae caused by DlScer\UAS.cJa expressed under the control of Scer\GAL4l(3)31-1-31-1 is strongly enhanced by Dp(1;2)51b or H1, and widespread transformation of microchaetae is also seen in these backgrounds. The transformation of the trichogen to a tormogen fate in the notal macrochaetae caused by DlScer\UAS.cJa expressed under the control of Scer\GAL4l(3)31-1-31-1 is enhanced by Df(3R)Dl-M2. The transformation of the trichogen to a tormogen fate in the notal macrochaetae caused by DlScer\UAS.cJa expressed under the control of Scer\GAL4l(3)31-1-31-1 is dominantly suppressed by Su(H)12, Df(3R)Espl3 or Tp(3;3)bxd110. Coexpression of NECN.Scer\UAS completely suppresses the transformation of the trichogen to a tormogen fate in the notal macrochaetae caused by DlScer\UAS.cJa expressed under the control of Scer\GAL4l(3)31-1-31-1. Coexpression of NECNΔ10-12.Scer\UAS fails to suppress the transformation of the trichogen to a tormogen fate in the notal macrochaetae caused by DlScer\UAS.cJa expressed under the control of Scer\GAL4l(3)31-1-31-1 in the presutural, anterior supraalar, anterior postalar, anterior scutellar and posterior scutellar bristles. Transformation of the trichogen to a tormogen fate in the anterior notopleural, posterior postalar and posterior dorsocentral bristles is slightly reduced in these flies. The bristle transformation phenotype seen when both SerScer\UAS.cGa and DlScer\UAS.cJa are expressed under the control of Scer\GAL4l(3)31-1-31-1 appears to be additive. The transformation of the trichogen to a tormogen fate in the notal macrochaetae caused by DlScer\UAS.cJa expressed under the control of Scer\GAL4l(3)31-1-31-1 is suppressed by co-expression of fngScer\UAS.cKa.
Coexpression of DlScer\UAS.cJa and SerBd.Scer\UAS.T:Hsap\MYC via Scer\GAL4Ser.PGF did not alter the extent of cell proliferation in the dorsal wing compartment in discs or adult wing blades. Coexpression of DlScer\UAS.cJa and SerBd.Scer\UAS.T:Hsap\MYC via Scer\GAL4ptc-559.1 leads to variable but consistent reduction in the extent of ventral compartment cell proliferation as compared to that produced by DlScer\UAS.cJa alone. The antimorphic effects of SerBd.Scer\UAS.T:Hsap\MYC (therefore SerBd-3) are compartmentally restricted in the wing disc.

However more than one aCC/pCC neuron pair can sometimes be seen per segment. When DlScer\UAS.cJa is driven by Scer\GAL4W381 in a Dl3 background, the cell fate specification phenotype is rescued, but the neurogenic phenotype is not.
