Wing disc clones expressing brk[Scer\UAS.cJa] under the control of Scer\GAL4[Scer\FRT.Act5C] grow poorly and are rarely recovered. Expression of brk[Scer\UAS.cJa] under the control of Scer\GAL4[GMR.PF] produces flies with small eyes compared to controls.
Expression of brk[Scer\UAS.cJa] using Scer\GAL4[salm-459.2] results in moderate wing size reduction and vein loss.
Somatic clones of brkScer\UAS.cJa; Scer\GAL4Act5C.Switch.PR cells in mid-third instar wing discs show autonomous decreases in numbers of cells in S-phase after 14 hours treatment with 20μg/ml RU486. However, if the edges of these clones are located medially they cause a large increase in the of cells in S-phase adjacent to the medially located edge of the clone. No increase in cell death occurs prior to 26 hours after the initiation of RU486 treatment. A small increase in apoptosis is seen at 48 hours.
brkScer\UAS.cJa; Scer\GAL4Bx-MS1096 flies have small wings in which some or all of the veins are shifted, truncated or completely missing. Wing veins L2 and L5 are particularly stringly affected. brkScer\UAS.cJa; Scer\GAL4C-765 flies have a range of venation phenotypes, including selective loss or displacement of L2 and L5 veins, a single central vein (L3) or complete loss of veins. In brkScer\UAS.cJa; Scer\GAL4vg.int2.1 flies the wing veins are crowded into the central region of the wing blade, close to the anterior-posterior compartment boundary.
brkScer\UAS.cJa; Scer\GAL4C-765 and brkScer\UAS.cJa; Scer\GAL4nub-AC-62 flies have reduced wings. The degree of size reduction in the wings of brkScer\UAS.cJa; Scer\GAL4C-765 flies varies with temperature - it is much more severe in animals raised at 25oC than 17oC. This is also apparent in discs - with severe size reduction being see in wing leg and haltere discs of third instar larvae raised at 29oC, and less in those raised at lower temperatures. There is a slight increase in the level of apoptosis in the wing pouch of late third instar brkScer\UAS.cJa; Scer\GAL4nub-AC-62 wing discs, but there is also a decrease in the rate of cell division in the pouch, as measured by BrdU incorporation or cell division marker expression. A similar decrease in proliferation, restricted to the posterior compartment, is seen in brkScer\UAS.cJa; Scer\GAL4hh-Gal4 wing discs.
Expression of brkScer\UAS.cJa under the control of Scer\GAL4twi.PG blocks crystal cell development. Expression of brkScer\UAS.cJa under the control of Scer\GAL4srp blocks crystal cell development.
When brkScer\UAS.cJa is driven throughout the mesoderm by Scer\GAL4how-24B and Scer\GAL4twi.PG, there is a consierable reduction in cardiogenesis.
Expression of brkScer\UAS.cJa under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 has no effect on the testes.
Clones in the wing disc expressing brkScer\UAS.cJa under the control of Scer\GAL4Ubx.PdC do not survive in the centre of the disc, surviving only in lateral regions.
Expression of brkScer\UAS.cJa under the control of Scer\GAL4Kr.PM results in a reduced number of neurons in the dorsal and lateral peripheral nervous system. Segments expressing brkScer\UAS.cJa show no increase in apoptosis compared to other abdominal segments that are not expressing brkScer\UAS.cJa.
When expression is governed by Scer\GAL4bi-omb-Gal4 the area of the wing blade is dramatically reduced, leaving the alula and costa largely unperturbed. Corresponding third larval instar discs are reduced and do not have the normal folded morphology.
Embryos expressing brkScer\UAS.cJa under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 have a completely ventralised cuticle.

Expression of yki[S168A.Scer\UAS.T:SV5\V5] partially suppresses the underproliferation seen when brk[Scer\UAS.cJa] is expressed in wing disc clones under the control of Scer\GAL4[Scer\FRT.Act5C]. Co-expression of tkv[Q253D.Scer\UAS.cNc] suppresses the growth defects further, producing some clone overgrowth, whilst the addition of Mad[Scer\UAS.T:Zzzz\FLAG] increases clone overgrowth even further. Co-expression of tkv[Q253D.Scer\UAS.cNc] and Mad[Scer\UAS.T:Zzzz\FLAG] does not suppress the growth and survival defects seen when brk[Scer\UAS.cJa] is expressed in wing disc clones under the control of Scer\GAL4[Scer\FRT.Act5C]. Co-expression of Mad[Scer\UAS.T:Zzzz\FLAG] and tkv[Q253D.Scer\UAS.cNc] suppresses the small eye phenotype seen when brk[Scer\UAS.cJa] is expressed under the control of Scer\GAL4[GMR.PF]. Expression of yki[S168A.Scer\UAS.T:SV5\V5] suppresses the small eye phenotype seen when brk[Scer\UAS.cJa] is expressed under the control of Scer\GAL4[GMR.PF]. When tkv[Q253D.Scer\UAS.cNc] is additionally expressed overgrown eyes are seen. Expression of ban[GS] suppresses the underproliferation seen when brk[Scer\UAS.cJa] is expressed in wing disc clones under the control of Scer\GAL4[Scer\FRT.Act5C]. The eye growth defects seen when brk[Scer\UAS.cJa] is expressed under the control of Scer\GAL4[GMR.PF] are also rescued.
pucScer\UAS.cMa does not rescue with development in brkScer\UAS.cJa; Scer\GAL4nub-AC-62 flies. BacA\p35Scer\UAS.cHa does not suppress the reduction of wing pouch size seen in brkScer\UAS.cJa; Scer\GAL4nub-AC-62 wing discs - even when 2 copies of BacA\p35Scer\UAS.cHa are present. The reduction in wing size seen in brkScer\UAS.cJa; Scer\GAL4nub-AC-62 flies is not suppressed by pucScer\UAS.cMa.
The addition of brkScer\UAS.cJa suppresses the ectopic eye phenotype seen in eyScer\UAS.cHa, Scer\GAL4bi-omb-Gal4 animals. The addition of brkScer\UAS.cJa suppresses the ectopic eye phenotype seen in eyScer\UAS.cHa, Scer\GAL4dpp.blk1 animals.
Co-expression of BacA\p35Scer\UAS.cHa allows the survival of clones expressing brkScer\UAS.cJa under the control of Scer\GAL4Ubx.PdC in the centre of the wing disc, although their proliferation is impaired. Co-expression of pucScer\UAS.cMa allows the survival of clones expressing brkScer\UAS.cJa under the control of Scer\GAL4Ubx.PdC in the centre of the wing disc.
