Description

GMR regulatory sequences drive expression of a dominant negative form of ebi.

The expression of the first 334 amino acids of ebi is governed by GMR regulatory sequences.

Detailed Description

After eclosion, ebi1-334.GMR/+ flies possess a near-normal complement of photoreceptors in each ommatidium. Five weeks later, however, these retinas are severely degenerated.

One copy of P{GMR-ebi.1-334} produces a slightly abnormal rough-eye phenotype, and two copies produces a severe rough-eye phenotype.

5 week old ebi1-334.GMR/+ photoreceptor cells in the eye show defective mitochondria with swollen and abnormal cristae.

Expression of ebi1-334.GMR causes a mild defect in eye morphology.

ebi1-334.GMR homozygotes have a mild rough eye phenotype.

ebi1-334.GMR causes a weak eye phenotype.

Additional Comments
Genetic Interactions
Statement
Reference

Df(2R)X1/+ or Jra1/+ suppresses the rough eye phenotype seen in flies with two copies of ebi1-334.GMR.

Df(3L)H99/+, Jra1/+, hepr75/+, but not hid05014/+, suppresses the late-onset retinal degeneration phenotype of ebi1-334.GMR/+ flies. Diap14/+ enhances this phenotype.

Expression of ebi1-334.GMR strongly enhances the eye morphology phenotype of Scer\GAL4GMR.PF>chnGS17605 and Scer\GAL4GMR.PF>chnGS11450 flies but fails to enhance the eye phenotype of Scer\GAL4GMR.PF>chnGS17892, Scer\GAL4GMR.PF>chnScer\UAS.cTa and Scer\GAL4GMR.PF>chnGS2112 flies.

The mild rough eye phenotype of ebi1-334.GMR homozygotes is almost completely suppressed by rno2/+ or rno5/+.

The combination of ebi1-334.GMR and Dl9P/+ leads to flies with a significantly rough eye phenotype.

Comments

Scer\GAL4GMR.PU-mediated expression of ebiScer\UAS.cTa rescues the retinal degeneration phenotype seen in 5-week old ebi1-334.GMR/+ flies.
