Construct: The 1.94kb EcoR1-Sca1 and the 1.04kb Spe1(filled in)-EcoR1 fragments of the 1A pbl cDNA are expressed under the control of Scer\UAS sequences. Amino acids 497-549, at the C terminal of the DH domain, are removed.

Expression of pblΔDH497-549.Scer\UAS under the control of Scer\GAL4GMR.PF results in an obvious roughening of the adult ommatidial array.
When pblΔDH497-549.Scer\UAS is driven by Scer\GAL4Bx-MS1096 a multiple wing hair phenotype is seen. Nuclei in the wing are heterogeneous in size and, and the multiple wing hair phenotype is a consequence of a failure in cytokinesis.
pblΔDH497-549.Scer\UAS; Scer\GAL4en-e16E flies have only subtle morphological defects in the posterior compartment of their wings.
Overexpression of pblΔDH497-549.Scer\UAS in the eye driven by Scer\GAL4GMR.PF, induces disruption of the ordered structure of the eye, with fusions of ommatidia, loss of many bristles, variation in numbers of rhabdomeres per ommatidium and decrease in the number of pigment cells. This phenotype is dominantly enhanced by mutations in pbl.

The addition of CycB32 suppresses the pblΔDH497-549.Scer\UAS wing hair phenotype in a dose dependant manner.
The eye phenotype caused by expression of pblΔDH497-549.Scer\UAS by Scer\GAL4GMR.PF is dominantly enhanced by Rho172R and dia2.

Comments
pblΔDH497-549.Scer\UAS; Scer\GAL4prd.RG1 fails to significantly rescue the formation of eve positive clusters of dorsal mesodermal cells in stage 11 pbl2/pbl3 embryos.
