Drives expression in Pdf and LNv neurons.

In larval brain, expression in four cells in each hemisphere; these persist into adulthood and become the small ventrolateral neurons. In adult brain, also expressed in four large ventrolateral neurons, which emerge during metamorphosis. In both larval and adult stages, expressed in a small number of cells in the posterior-most CNS.

Expression of PdfUAS.cRa with ScerGAL4P2.4.Pdf is weaker than wildtype Pdf expression, but expression is within the LNv neurons.

Activation of Scer\GAL4P2.4.Pdf-positive neurons (by the expression of TrpA1UAS.(B).cKa under the control of Scer\GAL4Lk.PM) leads to increased female fecundity.

Elimination of molecular clock function (using cycΔ.UAS.Tag:MYC) in Scer\GAL4P2.4.Pdf cells in adults significantly reduces the strength of the circadian rhythm of sleep-wake cycles, compared to controls.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4P2.4.Pdf cells significantly reduces the strength of the circadian rhythm of sleep-wake cycles, compared to controls.

Blocking clock function (using cycΔ.UAS.Tag:MYC) of Scer\GAL4P2.4.Pdf neurons significantly reduces the circadian rhythmicity of adult emergence after transfer to constant darkness, compared to controls.

Ablation of neurons expressing Scer\GAL4P2.4.Pdf has no effect on startle-induced climbing ability.

Optogenetic activation (using Crei\ChR2UAS.cSa with blue light activation) of Scer\GAL4P2.4.Pdf neurons in the larva results in excitatory junction potentials in anal sphincter muscles and strong contraction of anal sphincter muscles and other hindgut muscles, no excitation or contraction is seen due to blue light in controls.

Silencing (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4P2.4.Pdf neurons significantly increases the interval of defecation; when Scer\GAL4eve.RN2 neurons are additionally silenced, there is a significantly reduced frequency of defecation, compared to controls, with barely any hindgut movement.
