Overexpression of fjScer\UAS.cZa under the control of Scer\GAL4salm-459.2 does not affect wing hair polarity.

Expression of fjScer\UAS.cZa under the control of Scer\GAL4αTub84B.PL in clones in the dorsal adult abdomen results in the reversal of hairs in front of the clone.

Expression of fjScer\UAS.cZa under the control of Scer\GAL4bi-omb-Gal4 results in weak polarity reversals in the eye disc.

Expression of fjScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 results in loss of the anterior crossvein and subtle change sin wing hair polarity (wing hairs point towards the region overexpressing fjScer\UAS.cZa mainly in the distal part of the wing).

The wings of ftScer\UAS.cMa; Scer\GAL4en-e16E flies have planar polarity defects in much of the posterior compartment and a a posterior crossvein which has moved into distally, into line with the anterior crossvein. ftScer\UAS.cMa; Scer\GAL4salm-459.2 have a similarly misplace posterior cross-vein, with severe planar cell polarity defects clustered around it. ftScer\UAS.cMa; Scer\GAL4ap-md544 lack a posterior crossvein, have an incomplete anterior crossvein and scattered strong planar polarity defects. ftScer\UAS.cMa; Scer\GAL4ptc-559.1 flies also have a distally displaced posterior crossvein with planar polarity defects immediately proximal and distal to it.

Flies expressing fjScer\UAS.cZa under the control of Scer\GAL4αTub84B.PL in a fjd1/fjN7 background have a normal overall pattern of polarisation in the eye; an equator can easily be discerned. 92.9% of ommatidia are correctly constructed and rotated in these animals. Expression of fjScer\UAS.cZa under the control of either Scer\GAL4αTub84B.PL or Scer\GAL4fj-VG1 has little effect on ommatidial polarity, with only occasional mis-specified ommatidia being seen. Expression of fjScer\UAS.cZa under the control of Scer\GAL4bi-omb3-GAL4 has a profound effect on ommatidial polarity; near the dorsal pole, large-scale reversals of ommatidial polarity are seen. Expression of fjScer\UAS.cZa under the control of Scer\GAL4bi-omb3-GAL4 in a fjd1/fjN7 background has a profound effect on ommatidial polarity; near the dorsal pole, large-scale reversals of ommatidial polarity are seen.

fjScer\UAS.cZa; Scer\GAL4bi-omb-Gal4 flies have shorter wings with anterior and posterior crossveins closer together than wild-type. These flies have eyes with several rows of inverted ommatidia at their dorsal and ventral edges. fjScer\UAS.cZa; Scer\GAL4Act5C.PI somatic clones in the eye cause non-autonomous inversions of one or two rows of ommatidia on the equatorial side of the clone.

Within the anterior compartments of the adult abdominal cuticle, somatic clones in which fjScer\UAS.cZa is expressed under the control of the Scer\GAL4Ubx.PdC only exhibit planar polarity defects when they are located in the posterior of the compartment. Within the clone there are whorls, and these can extend outside the clone in the anterior, but not the posterior direction. Clones within the posterior compartment appear normal. Polarity defects result when fjScer\UAS.cZa is expressed under the control of the Scer\GAL4αTub84B.PL in somatic clones in the adult abdominal cuticle (generated by loss of Scer\GAL80αTub84B.PL through mitotic recombination): In the anterior compartment clones rarely have whorls, and the hairs are reversed within the anterior part of the clone. This reversal extends anterior to the clone itself by as much as 6 or 7 cell diameters. However, some clones within the extreme anterior portion of the hair producing territory of the anterior compartment cause little or no changes in polarity anterior to the clone. Clones in the posterior compartment are also associated with polarity reversals: hairs within the posterior half of the clone as well as hairs behind are reversed and now point anteriorly. When a clone in the anterior compartment abuts the posterior compartment boundary then non-autonomous reversal of hair polarity is seen both anterior and posterior to the clone.

Expression of fjScer\UAS.cZa under the control of Scer\GAL469B results in ectopic joint-like structures in the tarsal segments, loss of proximal-distal growth within the tarsal segments and loss of the joint between tarsal segments 2 and 3. Expression of fjScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 results in significant truncation and fusion of tarsal segments. Expression of fjScer\UAS.cZa under the control of Scer\GAL4dpp.blk1 results in leg truncations and fusion of tarsal segments, as well as cuticular abnormalities and occasional outgrowths from the leg. Some of these outgrowths appear to be segmented and most have at least some bristles. Expression of fjScer\UAS.cZa under the control of Scer\GAL4en-e16E results in leg truncations and outgrowths. Clones in the leg expressing fjScer\UAS.cZa under the control of Scer\GAL4Act5C.PI that span a segment border result in the fusion of most segments. In the tarsus, fusion of the tarsal segments is accompanied by reduced growth. Loss of joint structures is seen both within and adjacent to the clone. Occasional outgrowths from the leg are seen in legs containing larger clones. These outgrowths are composed entirely of wild-type tissue, with clone tissue being close to the outgrowth.

When fjScer\UAS.cZa is driven by Scer\GAL4Scer\FRT.Act5C occasional swirls in the wing hair pattern are seen in the proximal regions in the wing. The overall wing length is shortened, specifically the distal wing is shortened but the proximal wing size remains unaffected. These flies also have defects in the leg - including proximal-distal shortening and tarsal fusion. Scer\GAL4Scer\FRT.Act5C driven expression also produces wing vein phenotypes, including loss of the posterior crossvein and displacement of vein 4 at the junction with the wing margin as well as the deletion of the anterior cross vein in the region. When Scer\GAL4ptc-559.1 drives expression of fjScer\UAS.cZa in a stripe along the anterior margin of the anterior/posterior compartment boundary of the developing wing, nearby wing hairs rotate towards the highest levels of fj expression close to the compartment boundary. When Scer\GAL4ptc-559.1 drives expression of fjScer\UAS.cZa in the leg, extreme proximal-distal axis shortening and loss of distal structures, while the coxa, a proximal structure, appears largely unaffected.

When fjScer\UAS.cZa is driven at the poles of the eye by Scer\GAL4bi-omb-Gal4, dorsoventral inversions of ommatidial polarity are seen at poles of the eye, often with 3 or more rows inverted. When driven by Scer\GAL4Scer\FRT.Act5C ommatidial inversions are seen on the equatorial boundary of expression. When driven by Scer\GAL4ey.PH no phenotype is seen. When driven by Scer\GAL4hs.2sev or Scer\GAL4ey.PH occasional ommatidia are seen with inverted polarity relative to their neighbours. Tese phenotypes are similar but stronger than the homozygous mutant fj phenotypes.

When fjScer\UAS.cZa is overexpressed under the control of Scer\GAL4Bx-MS1096 in a pk30 mutant wing, the pk30 wing hair phenotype is modified to a more distal polarity in the anterior and distal posterior regions.

Overexpression of fjScer\UAS.cZa under the control of Scer\GAL4salm-459.2 in a pk30 mutant wings results in changes in wing hair polarity (relative to the wild-type pattern) outside of the main Scer\GAL4salm-459.2 expression domain.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing fjScer\UAS.cZa under the control of Scer\GAL4αTub84B.PL in a stan- background (the cells within the clone are stanE59/stanE59 while the cells surrounding the clone are stan3/stanE59) result in a reversal of cell polarity in cells anterior to the clone (hairs in front of the clone point forwards), as occurs in clones induced in a wild-type background.
