Drives expression in dopaminergic and serotonergic neurons, and their precursors.

ScerGAL4Ddc.PL drives expression very weakly in the ellipsoid and the fan-shaped bodies.

ScerGAL4Ddc.PL drives expression in a subset of the dopaminergic PPL2ab neurons and in the majority of the PAM neurons (includes MB-M3 and MB-MP1 neurons).

Expression observed in neurons that innervate the mushroom body horizontal lobes and the subsesophageal ganglion.

Expression in the midgut is found in a distinct group of Dh31-expressing enteroendocrine cells located at the junction region of the anterior midgut and the acid-secreting portion of the larval midgut. This junction region occurs at a conspicuous and characteristic U-shaped bend in the gut. These are referred to as the "midgut junction Dh31-expressing cells".

Expression is seen in dopaminergic and serotonergic neurons and their precursors, totalling 9-11 neurons per abdominal half-neuromere by larval stage L1. A precise pattern can be found in FBrf0160715.

Expression is observed in Ddc-expressing neurons.

Inhibition (using shits1.IVS.p10.20xUAS at 32[o]C) of Scer\GAL4Ddc.PL cells in adults significantly reduces 3-min associative olfactory water-reward memory, compared to controls; inhibition during acquisition alone also significantly reduces 3-hour and 24-hour memory.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 31[o]C) of Scer\GAL4Ddc.PL cells in thirsty or water-sated adults, in combination with a paired odor, results in a significant increase in 3-min, 3-hour and 24-hour associative attractive olfactory memory, compared to controls.

Inhibition (using Ctet\tetXTNT.UAS) of a subset of Scer\GAL4Ddc.PL cells (expression restricted using Scer\GAL80ple.PK) does not affect latency of escape from heat or associative aversive (heat) place memory, compared to controls; the increases in escape latency and aversive memory due to unexpected high (41[o]C) temperature pre-exposure are significantly lower than those of controls.

Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4Ddc.PL cells, in combination with a Gal80[ts] transgene to restrict expression to the adult stage, does not affect aversive temperature or shock olfactory memory, compared to controls, and there is no effect on temperature avoidance.

Inhibition (using shi1.UAS at 33[o]C) of Scer\GAL4Ddc.PL cells in adults significantly reduces appetitive, but not aversive, 1-min visual memory, compared to controls; inhibition during training alone, but not testing, significantly reduces appetitive, but not aversive, 30-min visual memory, compared to controls; inhibition does not significantly affect sugar preference, compared to controls.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of Scer\GAL4Ddc.PL cells in starved, but not fed, adults, in combination with a paired odor, results in a significant increase in 2-min associative attractive olfactory memory, compared to controls; this memory increase is still seen when activation is restricted to a smaller number of Scer\GAL4Ddc.PL cells using Scer\GAL80ple.PK, but not Scer\GAL80R58E02; a significant increase in 12-hour memory also occurs when flies are starved or fed prior to training, but not if they are fed immediately before testing.

Inhibition (using shi1.UAS at 32[o]C) of Scer\GAL4Ddc.PL cells in adults fed immediately before training does not significantly affect 3-hour associative appetitive olfactory memory, compared to controls; inhibition before and during training, but not after training and during testing, significantly reduces 30-min associative appetitive olfactory memory, compared to controls; inhibition also significantly reduces short-term associative appetitive olfactory memory, compared to controls, but not when restricted to a smaller subset of Scer\GAL4Ddc.PL cells using Scer\GAL80R58E02; inhibition has no effect on naive sugar preference, compared to controls.

Animals expressing two copies of Scer\GAL4Ddc.PL exhibit a greatly reduced lifespan, compared to wild-type. their locomotion remains normal.
