Description

Construct: Expression of p53 open reading frame is driven by the GMR regulatory sequences. An alpha tubulin 3' UTR provides stability to the message.

p53GMR.Ex induces a small eye.

Flies expressing p53GMR.Ex have a reduced eye, due to increased cell death.

Expression of p53GMR.Ex produces flies with small and rough eyes with a glossy appearance. Massive cell death is seen in the posterior half of the third instar larval eye disc and the numbers of R7 photoreceptor neurons and cone cells are strongly reduced compared with wild type. All other photoreceptor neurons appear largely normal. In pupal p53GMR.Ex eye disc ommatidia are severely misorganized, as indicated by enlarged interommatidial space, ommatidial fusions and reduced number of cone cells in each ommatidium.

Expression of p53GMR.Ex in the posterior portion of late larval eye discs is sufficient to induce apoptosis.

Expression of p53GMR.Ex induces apoptosis in the developing eye disc, which results in a dramatic reduction in adult eye size compared to wild type.

Flies expressing p53GMR.Ex have eye tissue with no evidence of intact ommatidia or bristles.

Adults are viable with small, rough eyes with fused ommatidia. This is due to widespread apoptosis in cells expressing p53GMR.Ex. Cells initiate apoptosis within 3-6 hours after p53GMR.Ex is expressed. p53GMR.Ex neither blocks nor delays the transition from G1 to S phase in the second mitotic wave in the eye disc. However the pattern of mitosis is affected with entry into M phase or duration of M phase affected.

Expression of CorpEY03495 under the control of Scer\GAL4GMR.PF suppresses the increased cell death and reduced eye phenotypes caused by expression of p53GMR.Ex.

Corp95B enhances the increased cell death and reduced eye phenotypes caused by expression of p53GMR.Ex.

Induction of ArkG8 third instar larval eyes disc clones blocks the cell death seen in p53GMR.Ex mutants.

The p53GMR.Ex-mediated induction of apoptosis in the posterior portion of late larval eye discs is reduced in hpo42-47, wtsx1 and sav3 clones.

The increased apoptosis in the eye disc caused by expression of p53GMR.Ex is suppressed in a NcΔA8/NcΔA8 background, but the reduction in adult eye size caused by p53GMR.Ex is not suppressed if the eye is made homozygous for NcΔA8 (using the eyFLP method).

Expression of Hsap\CDKN1AGMR.PH rescues the cell death and R7 photoreceptor loss seen in p53GMR.Ex third instar larval eye discs. The adult eye ablation phenotype is also rescued.
