Flies expressing perScer\UAS.cYa under the control of Scer\GAL4tim.PE in a Scer\GAL80Pdf.PS background show rhythmic locomotor activity under constant light conditions, in contrast to wild-type flies, which are uniformly arrhythmic under these conditions.
Flies expressing perScer\UAS.cYa, under the control of Scer\GAL4P0.5.Pdf, exhibit low levels of arrhythmicity (approximately 22.2% versus 13%) although the period length is increased by approximately 1 hour.
Flies mutant for per01 expressing perScer\UAS.cYa under the control of Scer\GAL4elav-C155 and repressed by Scer\GAL80Pdf.PS in Pdf expressing cells, become gradually arrhythmic under DD conditions. These flies however, manifest a wild-type-like anticipation of lights-on as well as lights off peak in LD.
When perScer\UAS.cYa is driven by Scer\GAL4cry.PP no induction of ectopic clocks is seen. A reduction is seen behavioural rhythmicity in DD conditions.
When expression is driven in a per+ background by Scer\GAL4elav-C155 or Scer\GAL4Act5C.PI, arythmicity occurs. Two copies of P{UAS-per.Y} give a stronger effect than one. The effect is weaker with Scer\GAL4tim.PE and not evident with Scer\GAL4P2.4.Pdf.

Expression of cyc[Scer\UAS.T:Hsim\VP16] under the control of Scer\GAL4[elav-C155] in a per[01], per[Scer\UAS.cYa] background only shortens the circadian locomotor activity period by 20 minutes, in contrast to the shortening of 2 hours seen when cyc[Scer\UAS.T:Hsim\VP16] is expressed under the control of Scer\GAL4[elav-C155] in a wild-type background.
Flies expressing perScer\UAS.cYa and cryScer\UAS.cEa, under the control of Scer\GAL4P0.5.Pdf, exhibit low levels of arrhythmicity (approximately 15.7% versus 13%).

Expression of per[Scer\UAS.cYa] in neuronal cells (under the control of Scer\GAL4[elav-C155]) or in circadian cells (under the control of Scer\GAL4[tim.PE]) rescues daytime temperature preference rhythms in per[01] mutant flies. Expression of per[Scer\UAS.cYa] in M cells, under the control of Scer\GAL4[P2.4.Pdf], fails to rescue daytime temperature preference rhythm under light-dark conditions, although a weaker-amplitude rhythm with an abnormal phase is present in these flies. Expression of per[Scer\UAS.cYa] in both M and E cells with Scer\GAL4[cry.PE] is unable to rescue daytime temperature preference rhythms under light-dark conditions. Expression of per[Scer\UAS.cYa] in the DN2 neurons (under the control of Scer\GAL4[Clk.int1-3]) rescues approximately 46% of the per[01] mutant phenotype for daytime temperature preference rhythm. Expression of per[Scer\UAS.cYa] in the DN2 neurons (under the control of Scer\GAL4[Clk.int1-3] and Scer\GAL80[Pdf.PS]) rescues approximately 66% of the per[01] mutant phenotype for daytime temperature preference rhythm. Expression of per[Scer\UAS.cYa] in DN2 neurons under the control of Scer\GAL4[Clk.int1-3] and Scer\GAL80[Pdf.PS] does not rescue the per[01] locomotor activity phenotypes under either light-dark or temperature cycles. These flies are also arrhythmic in dark-dark conditions.
Expression of per[Scer\UAS.cYa] under the control of Scer\GAL4[elav-C155] can rescue the arrhythmic locomotor activity phenotype of per[01] flies, with 65.2% of the rescued flies being rhythmic for locomotor activity.
The degree of rescue of the per01 locomotor rhythm defects depends to some extent on the site of insertion of the rescuing P{UAS-per.Y} transgene and is maximally >90% and minimally >50%.
