Description

Insertion 298bp upstream of the annotated garz transcription start site.

The orientation of the P{EP} element is the same as the direction of garz transcription.

The P{EP} element is inserted 295 nucleotides 5' to the predicted ATG start codon.

Lethal phase analysis of garzEP2028 homozygous mutants reveals early first instar larval lethality with a very low number of individuals surviving until the end of larval development.

Expression via Scer\GAL4fkh.PH results in a weakly penetrant but severe positioning ('hook') phenotype in the embryonic salivary gland.

Flies expressing garzEP2028 under the control of Scer\GAL4GMR.PF in the dark show normal rhabdomere development.

Flies expressing garzEP2028 under the control of Scer\GAL4GMR.PF and grown in bright light show some defects in rhabdomere structure and mild accumulation of endomembranes in the cell body of the photoreceptors.

Expression of garzEP2028 under the control of Scer\GAL4GMR.PF results in a very mild rough eye phenotype.

Mutant larvae expressing garzEP2028 under the control of Scer\GAL4elav-C155 have excessive or ectopic synapses. Mutant larvae expressing garzEP2028 under the control of Scer\GAL4elav-C155 have reduced or abnormal synapses.

In the ventral muscle region of mutant larvae expressing garzEP2028 under the control of Scer\GAL4elav-C155 ISN pathfinding defects can be seen. ISNb leaves the ISN at an abnormal point. It generates an abnormal axonal branch that splits into synaptic branches on incorrect muscles.

Co-expression of garzEP2028 strongly enhances the retina phenotype caused by expression of PldScer\UAS.cRa under the control of Scer\GAL4GMR.PF; only one or two poorly developed rhabdomeres are seen.

Expression of garzEP2028 under the control of Scer\GAL4GMR.PF in a CdsA1 background under bright light illumination results in significantly worse defects in rhabdomere size and structure compared to CdsA1 single mutants of flies expressing garzEP2028 under the control of Scer\GAL4GMR.PF in a wild-type background. There is no change in the accumulation of membranes in the cell body of the photoreceptors in the double mutant flies.

The rough eye phenotype caused by expression of Hsap\MAPTV337M.Scer\UAS under the control of Scer\GAL4GMR.PF is enhanced by garzEP2028.

Precise excision of the P{EP} element reverts the modifying effect of garzEP2028 on the rough eye phenotype caused by expression of Hsap\MAPTV337M.Scer\UAS under the control of Scer\GAL4GMR.PF.
