Expression of AlkScer\UAS.cLa under the control of Scer\GAL4sev.PU leads to severe defects in ommatidia formation in the anterior part of the adult eye.

Scer\GAL4sev.EP-mediated expression results in loss of ommatidia in the anterior region of the eye

Pan-neuronal expression of AlkScer\UAS.cLa under the control of Scer\GAL4elav-C155, and restricted to the adult stages using Scer\GAL80ts.αTub84B, causes significant deficits in associative olfactory learning compared to controls.

Expression of AlkScer\UAS.cLa under the control of Scer\GAL4Ras64B.PW, and restricted to the adult stages using Scer\GAL80ts.αTub84B, causes significant deficits in associative olfactory learning compared to controls. The wings of the resulting adult flies are also smaller than in wild type, due to reduced cell size rather than cell number.

Expression of AlkScer\UAS.cLa under the control of Scer\GAL4Alk.38, and restricted to the adult stages using Scer\GAL80ts.αTub84B, causes significant deficits in associative olfactory learning compared to controls.

Expression of AlkScer\UAS.cLa under the control of either Scer\GAL4Mef2.247 or Scer\GAL4c747, and restricted to the adult stages using Scer\GAL80ts.αTub84B, does not affect associative olfactory learning.

In early stage 11 AlkScer\UAS.cLa; Scer\GAL4twi.PG embryos, the visceral mesoderm contains fusion competent myoblasts, but no muscle founder cells. At later stages, visceral mesoderm derivatives appear to be absent from these embryos, and the somatic musculature is defective: Several muscles are small and display a spindle-like shape with long and thin projections, indicating that only few myoblasts fuse to form the muscles. In AlkScer\UAS.cLa; Scer\GAL4bap.3 embryos the muscle founder cells of the visceral mesoderm are present and may be increased in number.

When expression is driven by Scer\GAL4He.PZ, hemocyte proliferation is increased and lamellocyte numbers are increased though crystal cells are unaffected.

Large clones in the wing disc expressing AlkScer\UAS.cLa under the control of Scer\GAL4Act5C.PI result in disruption of disc development. Animals carrying clones expressing AlkScer\UAS.cLa under the control of Scer\GAL4Act5C.PI do not survive to adulthood.

The disrupted morphology of ommatidia in the anterior part of the adult eye of flies expressing AlkScer\UAS.cLa under the control of Scer\GAL4sev.PU is partially suppressed by co-expression of cnk5xAIR.Scer\UAS.T:Ivir\HA1 and even more strongly by co-expression of cnk5xAIR.Scer\UAS.T:Myr-Src42A.T:Ivir\HA1.

Expression of AlkScer\UAS.cLa under the control of Scer\GAL4elav-C155, Scer\GAL4Ras64B.PW or Scer\GAL4386Y in a Nf1E2 mutant background results in larval lethality. Larval lethality is also seen when AlkScer\UAS.cLa is expressed under the control of Scer\GAL4elav-C155 in a Nf1E2/+ background.

In combination with Scer\GAL80ts.αTub84B at restrictive 29[o]C, expression of AlkScer\UAS.cLa driven by Scer\GAL4386Y or Scer\GAL41471 fully rescues the increase of total sleep seen in Alkts/Alkts females; expression driven by Scer\GAL430Y or Scer\GAL4121Y leads to partial rescue and expression driven by Scer\GAL4007Y or Scer\GAL4MJ63 does not rescue the phenotype.
