Expression of Hsap\MAPTV337M.Scer\UAS under the control of Scer\GAL4C57 leads to significantly reduced density of perinuclear microtubules (MTs) in third instar larval muscles, with sparser and shorter fibers compared to controls. Scer\GAL4elav.PU-driven expression results in excess of satellite boutons at larval neuromuscular junctions.
Expression of Hsap\MAPTV337M.Scer\UAS driven pan-neuronally by Scer\GAL4elav-C155 leads to a significantly shortened lifespan compared to control flies; shortened lifespan is not as severe (significantly longer) than seen with expression of Hsap\MAPTScer\UAS.cWa or Hsap\MAPT[R406W.Scer\UAS]. Scer\GAL4elav-C155>Hsap\MAPTV337M.Scer\UAS flies show significant changes in the activity of antioxidant enzymes. Expression of Hsap\MAPTV337M.Scer\UAS driven in mushroom bodies by Scer\GAL4c739 leads to significant impairment in middle-term and long-term olfactory memory compared to controls.

The decreased density of perinuclear microtubules (MTs) in larval muscle cells characteristic for third instar larvae expressing Hsap\MAPTV337M.Scer\UAS (with the Scer\GAL4C57 driver) as well as the excess of satellite boutons at larval neuromuscular junctions (with the Scer\GAL4elav.PU driver) is fully suppressed by αTub84BK40Q homozygosity. Unexpectedly, the microtubule as well as the neuromuscular junction phenotype of Hsap\MAPTV337M.Scer\UAS-expressing larvae can be partially or fully rescued (respectively) by αTub84BK40R homozygosity too.
HDAC6KO and HDAC6KO/Df(1)ED7294 each rescue the defects in perinuclear microtubule density seen in the muscles of animals expressing Hsap\MAPTV337M.Scer\UAS under the control of Scer\GAL4C57. HDAC6KO completely suppresses the increase in the number of satellite boutons at the neuromuscular junction (NMJ) and the decrease in average NMJ length caused by expression of Hsap\MAPTV337M.Scer\UAS under the control of Scer\GAL4elav.PU.
The moderate rough eye phenotype and reduced adult eye size characteristic for flies expressing Scer\GAL4GMR.PU under the control of Hsap\MAPTV337M.Scer\UAS is suppressed by co-expression of any of: Glut1d05758, sliScer\UAS.cBa or fneKK108902, restoring a near wild-type phenotype, and significantly worsened by co-expression of any of the following: Glut1GD4552 , sliGD5822, β-SpecdsRNA.Scer\UAS, Hs6stGD3078, dlg1GD4689 (each expressed together with UAS-Dicer2 to enhance RNAi efficiency) or fneScer\UAS.cCa, exacerbating the rough appearance.
