E2f276Q1/E2f2329 Mpp6+t4 animals have a pattern of irradiation induced apoptosis in the late third instar wing disc which is identical to that seen in wild-type animals. (Mpp6+t4 is present to rescued loss of Mpp6 in Df(2L)E2f2329.
Nearly all follicle cells in stage 13 E2f2[329]/E2f2[1-188] egg chambers show a phenotype of ectopic genomic replication, as assayed by BrdU incorporation. This phenotype is fully penetrant.
10% of eggs laid by E2f2329 females (Df(2L)E2f2329 homozygotes with Mpp6 function rescued by Mpp6+t4) hatch. 97% of the eggs laid by E2f2329 females have chorion defects. The chorions are more translucent than normal, indicating that the eggshell is thin. In stage 10B through 13 egg chambers of E2f2329 females (Df(2L)E2f2329 homozygotes with Mpp6 function rescued by Mpp6+t4), BrdU incorporation is seen throughout the entire follicle cell nuclei, rather than in characteristic subnuclear foci (which is seen in wild type). The follicle cells do not undergo an additional endocycle in these females. The number of BrdU positive follicle cell nuclei in pre-stage 10B egg chambers of these females is similar to wild type. E2f2329 males (Df(2L)E2f2329 homozygotes with Mpp6 function rescued by Mpp6+t4) are fully fertile. 8% of eggs laid by E2f2329/Df(2L)DS8 females carrying E2f21-188 (Df(2L)E2f2329/Df(2L)DS8 females carrying E2f21-188 with Mpp6 function rescued by Mpp6+t1-188) hatch. 93% of the eggs laid by these females have chorion defects. In stage 10B through 13 egg chambers of E2f2329/Df(2L)DS8 females carrying E2f21-188 (Df(2L)E2f2329/Df(2L)DS8 females carrying E2f21-188 with Mpp6 function rescued by Mpp6+t1-188), BrdU incorporation is seen throughout the entire follicle cell nuclei, rather than in characteristic subnuclear foci (which is seen in wild type). The follicle cells do not undergo an additional endocycle in these females.

The ectopic genomic replication that is seen in the follicle cells of stage 13 E2f2[329]/E2f2[1-188] egg chambers is partially suppressed by Orc2[γ3]/+ or Mcm2[rL074]/+; there is an approximate 65% reduction in the number of follicle cells within an egg chamber that display ectopic genomic replication and there is an apparent reduction in the intensity of ectopic BrdU incorporation. The ectopic genomic replication that is seen in the follicle cells of stage 13 E2f2[329]/E2f2[1-188] egg chambers is partially suppressed by Orc5[1]; the phenotype is suppressed in 36% of egg chambers. The ectopic genomic replication that is seen in the follicle cells of stage 13 E2f2[329]/E2f2[1-188] egg chambers is partially suppressed by E2f[91]/+; 55% of the double mutant egg chambers show a 50% reduction in the number of follicle cells exhibiting ectopic genomic replication compared to the single mutant egg chambers.
