Reference

Homozygous males lack mature sperm in the seminal vesicle. Spermatocyte development occurs normally through all the mitotic divisions and both rounds of meiosis. Nuclear elongation and axoneme development are grossly normal. However, syncytial spermatid bundles fail to fully individualise, resulting in non-motile, decomposing spermatid cysts. The actin rich individualization complexes show asynchronous migration during individualization.

blanksKG00804 homozygous females crossed to wild type males produce a similar number of offspring as controls. However, no progeny are produced when homozygous blanksKG00804 males, or blanksKG00804/Df(3L)Exel6105 males are crossed to wild type females.

No mature sperm are present in seminal vesicles from blanksKG00804 homozygous males.

Early spermatids from blanksKG00804 mutants are indistinguishable from wild type. However, defects are seen in spermatid individualization: mutant testes contain syncitial spermatid bundles with irregular bulges and 'lumps' along the length of the tail clusters. During the elongation phase, blanksKG00804 spermatid nuclei alignment is disrupted, and the nuclei are scattered and out of register. DNA condensation appears unaffected, as judged by P{ProtA-EGFP.Z} expression. The initial formation of the individualization complex (IC) also appears normal in the mutant. However, the ICs fail to translocate in a coordinated fashion and they become misaligned with stalled bundle migrations.
