Eyes of Hsap\MAPTGMR.Ex.PJ flies have a rough surface.
Expression of Hsap\MAPTGMR.Ex.PJ leads to rough eye phenotype and formation of giant autophagic bodies in the adult retina along with accumulation of autophagic intermediates both in the third instar eye discs and adult retinas.
Expression of Hsap\MAPTGMR.Ex.PJ results in rough, reduced eyes, with the posterior area of the eye remaining more normal. Microtubule shafts appear disorganised in the retinas of flies expressing Hsap\MAPTGMR.Ex.PJ.
The eyes of flies expressing Hsap\MAPTGMR.Ex.PJ under the control of Scer\GAL4GMR.PU show moderate toxicity compared to controls, characterized by the loss of photoreceptor neurons, leading to a rough eye phenotype in adults.
Hsap\MAPTGMR.Ex.PJ adult eyes display a reduced, rough-eye phenotype, with fused and disordered ommatidia with missing and irregular bristles. Retinal sections show marked ommatidial disorganization and cell loss.
Flies carrying one copy of Hsap\MAPTGMR.Ex.PJ have a mild rough eye phenotype, with mild loss and disorganisation of photoreceptor neurons.
Flies that express one copy of Hsap\MAPTGMR.Ex.PJ display no obvious rough eye phenotype.
Mutants exhibit an abnormal eye phenotype on eclosion. They are reduced in size and display a rough phenotype in anterior regions, while posterior aspects appear normal. Ommatidia are fused and disordered, with missing, irregular, and occasional supernumerary mechanosensory bristles. Flies with two copies of the P{GMR-htau.Ex} insert have severely reduced eyes. In third instar larval eye discs expressing two copies of P{GMR-htau.Ex}, early cell loss, and irregularities in differentiating neurons are apparent. Loss of photoreceptor neurons and disorganisation of the normal ommatidial architecture is also observed. Vacuolisation is also seen in the medulla.

When Scer\GAL4GMR.PU>dcoK38R.Scer\UAS.T:Hsap\MYC is expressed along with Hsap\MAPTGMR.Ex.PJ, there is significantly increased disruption in the eye. The enhanced disruption of the eye is manifested by the appearance of melanized patches in the eye and a decreased average surface area of the eye.
The formation of giant autophagic bodies in the retina of adult flies expressing Hsap\MAPTGMR.Ex.PJ is partially suppressed by combination with a single copy of Atg1Δ3D or by Scer\GAL4GMR.PU-driven co-expression of either Rab5S43N.Scer\UAS or Rab7T22N.Scer\UAS.P\T.T:Avic\GFP-YFP but not Rab14S49N.Scer\UAS.P\T.T:Avic\GFP-YFP. Flies that express Hsap\MAPTGMR.Ex.PJ in spinΔ2b/+ mosaic eyes show a mild rough eye phenotype while flies that express Hsap\MAPTGMR.Ex.PJ in spinΔ2b homozygous mosaic eyes show a severe rough eye phenotype.
When sgg1 is added to Hsap\MAPTGMR.Ex.PJ, the eye phenotype is ameliorated. This is seen when one copy of the P{GMR-htau.Ex} insertion is added, but is very marked when 2 copies are present. When sggScer\UAS.cSa (when driven by Scer\GAL4elav-C155) is added to Hsap\MAPTGMR.Ex.PJ, the eye phenotype is enhanced. The eye is markedly reduced and there is an almost complete loss of normal ommatidia and a loss of the majority of bristles. Abundant discrete flam-shaped and fibrillar aggregates and vacuolisation are observed throughout the retina and underlying lamina and medulla. The addition of arm1 or arm4 suppresses the eye phenotype seen in Hsap\MAPTGMR.Ex.PJ animals almost to wild-type, except for the anterior margin. Hsap\MAPTGMR.Ex.PJ and armS10.Scer\UAS.T:Hsap\MYC (driven by Scer\GAL4hs.2sev) enhance each other leading to markedly reduced eye, with a complete loss of ommatidial borders and bristles. The addition of pan2 suppresses the eye phenotype seen in Hsap\MAPTGMR.Ex.PJ subtly. Hsap\MAPTGMR.Ex.PJ and panScer\UAS.cWa (driven by Scer\GAL4hs.2sev) enhance each other leading to more roughened anterior part of the eye, with fusion, pitting, and collapse of ommatidia along with irregularity of bristles. The addition of BacA\p35GMR.PH, Iap2GMR.PH or thGMR.PH suppresses the eye phenotype in Hsap\MAPTGMR.Ex.PJ animals.
