The addition of E2f2dsRNA.cSa dsRNA to SL2 cells treated with E2fdsRNA.cFa dsRNA allows the E2f2-deficient cells to enter S phase, but does not rescue the cell cycle phasing defects. Although some SL2 cells treated with both E2fdsRNA.cFa and E2f2dsRNA.cSa dsRNAs can enter S phase, these cells fail to proliferate. No increase in apoptosis is evident, suggesting that this failure to proliferate is not due to cell death. The proportion of G2-phase cells remains low, indicating that few of the BrdU-positive cells fully complete S phase. SL2 cells treated with E2fdsRNA.cFa, E2f2dsRNA.cSa and dapdsRNA.cFa dsRNAs show a similar cell cycle distribution as wild-type cells and these cell cultures increase in number, although at a rate that is approximately half that of control cells.
