Description

UASp regulatory sequences drive expression of pav, with mutations in putative nuclear localization sequences 4-7 (amino acid replacements K770A, N771A, R772A, R843A, K844A and R845A), tagged at the N-terminal end with mGFP6.

When pavNLS4-7*.Scer\UAS.P\T.T:Avic\GFP-m6 is driven by Scer\GAL4OK107 no gross morphological changes in the mushroom body axons is seen.

Egg chambers of females expressing pavNLS4-7*.Scer\UAS.P\T.T:Avic\GFP-m6 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 contain a dense filamentous network of microtubules in the cytoplasm and at the cortex of nurse cells as well as in ring canals. The egg chamber develops a single oocyte, but it becomes mispositioned. Egg chambers containing 32 cells are often seen in the ovaries of females expressing pavNLS4-7*.Scer\UAS.P\T.T:Avic\GFP-m6 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16. These 32-cell cysts each have two independent oocytes each with 4 ring canals, indicating that they are produced by the fusion of two egg chambers. In some cases all the nurse cells within the 32-cell cyst are similar in size, although there are some cases where 32-cell cysts contain fused egg chambers of different ages. The collapse of ring canals and breakdown of the actin skeleton between nurse cells is seen.

Co-expression of pavNLS4-7*.Scer\UAS.P\T.T:Avic\GFP-m6 driven by Scer\GAL4arm.PS is unable to block the cell-fate transformation induced by armS10.Scer\UAS.T:Hsap\MYC expression.

When both tumScer\UAS.T:Hsap\MYC,T:Avic\GFP-EYFP and pavNLS4-7*.Scer\UAS.P\T.T:Avic\GFP-m6 are driven in the mushroom body by Scer\GAL4OK107, a high occurrence of axon misguidance is seen.

Comments

Expression of pavNLS4-7*.Scer\UAS.P\T.T:Avic\GFP-m6 driven by Scer\GAL4e22c fails to rescue the epidermal pattern defects of pavB200 mutants.
