The FACS profile of S2 cells treated with RacGAP50CdsRNA.1110-1961 dsRNA for 72 hours differs from the profile of control cells; there is a clear increase in the 8C peak at the expense of the 2C and 4C peaks, suggesting that a substantial fraction of the cells have become polyploid. A high frequency of binucleated cells is seen. Together, these analyses indicate a frequent failure of cytokinesis in the dsRNA treated S2 cells. S2 cell cultures treated with RacGAP50CdsRNA.1110-1961 dsRNA show a slight increase in the number of apoptotic cells compared to control cultures. Metaphase and anaphase figures are morphologically normal in cultures of S2 cells treated with RacGAP50CdsRNA.1110-1961 dsRNA for 72 hours and the frequencies of anaphases relative to metaphases are comparable with those of untreated controls. However, the telophases are severely affected; many telophases have a shorter pole-to-pole length than normal (morphologically normal telophases occur at very low frequencies). These "short" telophases have typical telophase nuclei with fully decondensed chromosomes, but they lack the central spindle. A uniform F actin distribution is seen in 20-30% of the short telophases, whereas in the remaining short telophases F actin concentrates in a wide equatorial band.
