Stage 14 egg chambers from females expressing TimpUAS.cPa under the control of Scer\GAL4GMR47A04 show mostly an intact posterior basement membrane, instead of the mostly open posterior basement membrane observed in controls.

Expression of TimpScer\UAS.cPa under the control of Scer\GAL4prd.RG1 in embryos leads to heart development defects, with cardioblasts failing to extend apical protrusions and failing to form a lumen; misaligned and rounded cardioblasts are also observed, in contrast to wild type.

Expression of TimpScer\UAS.cPa driven by Scer\GAL4Act.PU during adulthood (using Scer\GAL80ts.αTub84B) causes a significant reduction in egg laying, there is a significant increase in mature follicle cells per female, and average time to ovulate significantly increases compared to wild type. The number of eggs laid in 6 hours after mating significantly decreases in Scer\GAL4Act.PU>TimpScer\UAS.cPa (with Scer\GAL80ts.αTub84B) females.

Expression of TimpScer\UAS.cPa driven by Scer\GAL4GMR47A04 (along with UAS-dcr2) leads to a significant decrease in egg laying and a significant increase in ovulation time in females, compared to controls.

Scer\GAL4repo.PU-mediated expression of TimpScer\UAS.cPa results an elongated ventral nerve cord in third instars.

Expression of TimpScer\UAS.cPa under the simultaneous control of both Scer\GAL4bab1-Agal4-5 and Scer\GAL4ptc-559.1 (using the temperature sensitive Scer\GAL80ts.αTub84B allele to limit expression to the adult stage) results in an increase in the average number of cells in the interfollicle stalks compared to wild type.

Flies expressing TimpScer\UAS.cPa under the control of Scer\GAL4da.G32 exhibit clear tracheal deformation. ~85 of trachea exhibit at least one prominent tracheal break.

Expression of TimpScer\UAS.cPa under the control of Scer\GAL4btl.PS (by inactivating Scer\GAL80ts.alphaTub84B through heat-shock), severely impairs air sac extension and results in a deformed structure. Instead of a single well-defined tip, air sacs display multiple tips.

Randomly generated mutant cells expressing TimpScer\UAS.cPa under the control of Scer\GAL4btl.PS rarely populate the tip-cell territory in the air sac primordium.

61% of third instar larvae expressing TimpScer\UAS.cPa under the control of Scer\GAL4btl.PS display broken tracheal dorsal trunks. 63% survive to pupariation.

Expression of TimpScer\UAS.cPa under the control of Scer\GAL4btl.PS perturbs imaginal disc:tracheal association and dorsal air sac primordium growth associated with the wing disc. Extension and growth of the dorsal air sac primordia is stunted in these mutants, and the developing air sac tubes are filled with polarised epithelial cells. The adult flies that develop from these larvae lack normal dorsal air sacs.

Expression of TimpScer\UAS.cPa under the control of Scer\GAL4ap-md544 results in ectopic basal lamina formation around the tunneled tracheae associated with the wing disc. The depth at which the disc-associated transverse connective and dorsal air sac position within the imaginal disc is altered in these animals. Tunneled trachea and dorsal air sac primordia also show ectopic extrusions from the plane of the imaginal disc. Extension and growth of the dorsal air sac primordia is stunted in these mutants, and the developing air sac tubes are filled with polarised epithelial cells.

Scer\GAL4elav.PU- or Scer\GAL4repo.PU-mediated expression of TimpScer\UAS.cPa results in aberrant ISNb and SNa morphology - defasciculation with loose bundling and ectopic branches.

Eye/antennal disc clones that express TimpScer\UAS.cPa under the control of Scer\GAL4Act5C.PI have a normal appearance.

Overexpression of TimpScer\UAS.cPa in C4da neurons under the regulation of Scer\GAL4ppk.1.9 results in larval dendrites remaining in the extracellular space at 20 hours after pupal formation.

Overexpression of TimpScer\UAS.cPa in C4da neurons under the regulation of Scer\GAL4ppk.PG results in larval dendrites remaining in the extracellular space at 20 hours after pupal formation.

Expression of TimpScer\UAS.cPa in border cells, driven by Scer\GAL4slbo.2.6 does not affect the formation of the egg chamber apical cap or the migration of the border cell cluster to the oocyte.

Scer\GAL4btl.PS; TimpScer\UAS.cPa third instar larva have broken dorsal trunks, frequently with black melanization at the anterior tip of the broken trachea. However, 94% (n = 69) of these animals have normal posterior spiracles. 88% of Scer\GAL4btl.PS; TimpScer\UAS.cPa animals survive to third instar, 46% pupariate, 35% undergo head eversion, 33% develop head bristles, but only 7% eclose. 92% of Scer\GAL4dpp.PH; TimpScer\UAS.cPa animals survive to third instar, but only 31% pupariate, and none of these develops any further.

The disrupted formation of the presumptive posterior crossvein (assessed by p-Mad staining) in the pupal wing observed in animals expressing Gyc76CVDRC.cUa under the control of Scer\GAL4hh.PU is rescued by co-expression of TimpScer\UAS.cPa but it also leads to aberrant accumulations of the extracellular matrix.

Expression of TimpScer\UAS.cPa in enteroblasts under the control of Scer\GAL4Su(H).GBE reduces tumor formation in the anterior midgut of Sox21a6 mutant flies.

The co-expression of TimpScer\UAS.cPa does not suppressed the long distance cell migration and scattering, and the basal delamination of cells resulting from the expression of biScer\UAS.cBa under the control of Scer\GAL4dpp.PU and Scer\GAL4C-765, respectively.

Co-expression of TimpScer\UAS.cPa suppresses the fused egg chamber phenotype seen in females expressing Mmp2Scer\UAS.cPa under the control of Scer\GAL4C587 (using the temperature sensitive Scer\GAL80ts.αTub84B allele to limit expression to the adult stage) and restores stalk cell numbers to normal.

Co-expression of TimpScer\UAS.cPa partially suppresses the dissemination of hindgut cells seen in flies expressing Ras85DV12.Scer\UAS under the control of Scer\GAL4byn-Gal4.

The cell invasion phenotype seen in cells expressing sds22GD11788 under the control of Scer\GAL4ptc-559.1 along the anterior-posterior compartment boundary of the wing disc is largely suppressed if the cells are also co-expressing TimpScer\UAS.cPa, but the abnormal apical folding phenotype is not suppressed.

Expression of TimpScer\UAS.cPa under the control of Scer\GAL4da.G32 suppresses the NMJ defects seen in Fmr1Δ50M mutant third instar larvae. The mutant over-branching and supernumerary bouton formation have been eliminated, and the accumulation of developmentally arrested satellite boutons is prevented.

Flies expressing TimpScer\UAS.cPa under the control of Scer\GAL4da.G32 in a Fmr1Δ50M mutant background are robust, healthy and fully viable.

Homozygous Fmr1Δ50M fully suppresses the tracheal defects seen when TimpScer\UAS.cPa is expressed under the control of Scer\GAL4da.G32.

Expression of Mmp2Scer\UAS.cPa in third instar larvae expressing TimpScer\UAS.cPa under the control of Scer\GAL4btl.PS (by inactivating Scer\GAL80ts.alphaTub84B through heat-shock), largely reverts the air sac primordium defects observed when TimpScer\UAS.cPa is expressed alone.

Scer\GAL4Act5C.PI>Ras85DV12.Scer\UAS, TimpScer\UAS.cPa, scrib1 eye/antennal disc clones produce a tumor mass larger than that seen in Scer\GAL4Act5C.PI>Ras85DV12.Scer\UAS, scrib1 disc clones. However, the tumor cell invasiveness observed in Scer\GAL4Act5C.PI>Ras85DV12.Scer\UAS, scrib1 clones is suppressed by expression of TimpScer\UAS.cPa.

Loss of cell adhesion observed in induced-NijA1-161.EC1.Mtn.T:Ivir\HA1 cells is partially-suppressed through expression of TimpScer\UAS.cPa under the control of Scer\GAL4MtnA.PK.

Co-expression of TimpScer\UAS.cPa, under the control of Scer\GAL4MtnA.PK, with NijAA.MtnA.T:Ivir\HA1 suppresses the loss of cellular adherence observed after NijAA.MtnA.T:Ivir\HA1 inducement.

The co-expression of TimpScer\UAS.cPa does not significantly suppress the long distance cell migration and scattering, and the basal delamination of cells resulting from the expression of Hsap\TBX2Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4dpp.PU and Scer\GAL4C-765, respectively.

Comments

The increased number of neuromuscular junction boutons in TimpS1 mutant third instar larvae is somewhat decreased by Scer\GAL4how-24B-driven expression of TimpUAS.cPa, but the expression is not sufficient to rescue the slow and irregular peristaltic contractions characteristic for the mutant larvae.
