Co-expression of Sp1[dsRNA.Scer\UAS], Sp1[GD4097] and Dcr-2[Scer\UAS.cDa] under the control of Scer\GAL4[dac-p7d23] (restricted to second instar stage onwards using Scer\GAL80[ts.αTub84B]) results in a strong reduction in the size of the medial part of the leg. Co-expression of Sp1[dsRNA.Scer\UAS], Sp1[GD4097] and Dcr-2[Scer\UAS.cDa] under the control of Scer\GAL4[Dll-em212] (restricted to second instar stage onwards using Scer\GAL80[ts.αTub84B]) results in a strong reduction in the size of the distal part of the leg. Co-expression of Sp1[dsRNA.Scer\UAS], Sp1[GD4097] and Dcr-2[Scer\UAS.cDa] under control of Scer\GAL4[btd-MD808] (restricted to second instar stage onwards using Scer\GAL80[ts.αTub84B]) results in loss of the antennal a1 and a2 segments. Co-expression of Sp1[dsRNA.Scer\UAS], Sp1[GD4097] and Dcr-2[Scer\UAS.cDa] under control of Scer\GAL4[btd-MD808] from embryogenesis onwards results in a reduction in the size of the entire leg, from coxa to the distal tip.

When Sp1dsRNA.Scer\UAS is expressed in a btdMD808 background (driven by the Scer\GAL4btd-MD808 aspect of the P{GawB}btdMD808 insertion), at most some reduction in the tarsus is seen in some mutant legs. When both btddsRNA.Scer\UAS and Sp1dsRNA.Scer\UAS are expressed in a btdMD808 background (at 29oC) and driven by the Scer\GAL4btd-MD808 aspect of the P{GawB}btdMD808 insertion), the animals are unable to hatch and the antennae and legs are affected in all cases, though to different extents. Segments I and II of the tarsus are often reduced. Leg segments are reduced in size and often fused. When these flies are grown at 17oC almost no alteration is seen.
