Description

UAS regulatory sequences drive expression of mature SREBP.

SREBP sequences encoding a constitutively active protein variant truncated right before the first transmembrane domain were cloned into P{Express-UAS}. The C terminal sequence is RGMATNSR. The variant protein is localized to the nucleus constitutively and is constitutively active.

Expression of an active form of SREBP in enterocytes, through expression of SREBPCdel.Scer\UAS under the control of Scer\GAL4Myo31DF-NP0001 results in an increase in intestinal lipid production and whole-body triglyceride storage.

Expression of HLH106Cdel.Scer\UAS under the control of Scer\GAL4HCH.Hand results in a reduction in diastolic and systolic diameters and a reduction in the average heart period length in the adult heart compared to controls. Upon being subjected to pacing-induced stress, hearts expressing HLH106Cdel.Scer\UAS under the control of Scer\GAL4HCH.Hand show higher rates of heart failure compared to control hearts at young ages (1 week).

Expression of HLH106Cdel.Scer\UAS under the control of Scer\GAL4HCH.Hand causes significant loss of longitudinal fibres and disarrayed inner transverse myofibrils in the adult heart.

Expression of HLH106Cdel.Scer\UAS under the control of Scer\GAL4Bx-MS1096 results in significant lethality at 25[o]C. At 18[o]C, flies with a severe deformation of the wing are seen.
