incf00285 mutant third instar larvae show increased number of boutons on neuromuscular junction. Postsynaptic muscle recordings reveal altered synaptic transmission in inc1/incf00285 larvae: the amplitude of spontaneous miniature postsynaptic potentials is comparable to controls but their frequency is reduced, the amplitude of evoked potentials is decreased as is the quantal content.

Mutant flies show a reduction in sleep duration compared to controls. Sleep is reduced during both light and dark periods, but the sleep levels of the mutant flies drop in anticipation of light-dark and dark-light conditions, suggesting that the circadian clock is intact. The mutant flies show a reduction in sleep bout length accompanied by an increase in sleep bout number and show an increased latency to sleep after lights-off. The mutant flies show a modest reduction in activity during wakefulness. The mutant flies show reduced sleep homeostasis, lacking a detectable sleep rebound after 12 or 24 hours of sleep deprivation. The mutant flies are more easily woken by a rotational stimulus compared to wild type controls.

A proportion of mutant flies lack either an α- or β-lobe in the mushroom body.

These homozygotes are also short lived compared to controls.

Expression of inc under the control of either Scer\GAL4inc.PP or Scer\GAL4Cha.7.4 from the Scer\UAS regulatory sequences present in the PBac{WH}incf00285 insertion rescue the defects in sleep duration seen in incf00285 mutants, while expression under the control of either Scer\GAL430Y, Scer\GAL4c309 or Scer\GAL4G0451 partially rescues the sleep duration defects. Expression under the control of Scer\GAL4Mef2.247 does not rescue the sleep duration defects. Expression under the control of either Scer\GAL430Y or Scer\GAL4Cha.7.4 rescues the sleep homeostasis defects of incf00285 mutants.

Expression of inc under the control of Scer\GAL4elav.Switch.PO during development (using RU486 to control the timing of expression) from the Scer\UAS regulatory sequences present in the PBac{WH}incf00285 insertion partially rescues the sleep defects of incf00285 mutants, but expression under the control of Scer\GAL4elav.Switch.PO during the adult stage alone does not result in significant rescue of the sleep defects.

DATfmn incf00285 double mutants do not sleep significantly less than either single mutant.

Expression of inc from the PBac{WH}incf00285 insertion (using either Scer\GAL4Act5C.PI or Scer\GAL4αTub84B.PL) completely rescues the reduction in sleep per day which is seen in incf00285 homozygotes.

Expression of inc from the PBac{WH}incf00285 insertion using any one of the following Scer\GAL4 drivers partially rescues the reduction in sleep per day which is seen in incf00285 homozygotes: Scer\GAL4n-syb.PS, Scer\GAL4qvr.PW, Scer\GAL4Cha.7.4, Scer\GAL4D42, Scer\GAL4inc.PS, Scer\GAL4VGlut.PS, Scer\GAL4c309 and Scer\GAL4cenB1A-Mai301.
