Expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PU does not cause any obvious eye defects in adults, or defects of axon projections from retinal neurons in third instar eye discs, as compared to wild type.

Expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PF at 29[o]C results in a small rough eye phenotype.

Third instar larvae expressing nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4Pxn.PS contain an increased proportion of lamellocytes as a proportion of total hemocyte number (5.5% compared to 0.5% in controls). Hemocytes are able to undergo phagocytosis; as in wild type they are able to engulf India ink particles.

Expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4bbg-C96 results in a wing notching phenotype.

Overexpression of nejScer\UAS.T:SV5\V5 driven by Scer\GAL4hs.PB does not affect hydroxyurea-mediated cell cycle arrest.

Expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PF results in severe degeneration of the retina.

Expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PF results in adults with a severe rough eye phenotype. Very few, if any, photoreceptor cells are present in newly eclosed adults, although the photoreceptor cells appear to be recruited and specified correctly within the eye imaginal disc in larvae. Cone cell formation during pupal development appears to be abnormal in these animals; the number of cone cells per ommatidium is increased to 5 or 6 cells (compared to 4 in wild type). The severe rough eye phenotype caused by expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PF is suppressed if the flies also carry nej3/+.

Flies expressing nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PF have smooth eyes which completely lack ommatidia and mechanosensory bristles. Photoreceptors are not detected in these animals.

Co-expression of E(z)c.hs partially suppresses the mutant eye phenotype caused by expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4GMR.PF at 29[o]C; the eyes are normal in size and the rough eye phenotype is much milder.

100% of Pc3/+ males that are also expressing nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4hs.PB at 29[o]C have sex comb bristles on the T3 legs.

Expression of nejScer\UAS.T:SV5\V5 under the control of Scer\GAL4ey.PH suppresses the no-eye phenotype of soD/+; eye imaginal discs are nearly normal in size and contain large numbers of photoreceptor cell clusters, and in adults, the eyes are fully pigmented, although they are not normally patterned.

Xenogenetic Interactions
Statement
Reference

Co-expression of nejScer\UAS.T:SV5\V5 partially rescues the small eye and lens fusion phenotype seen in adult flies expressing Hsap\APPAβ42.Scer\UAS.cUa under the control of Scer\GAL4GMR.PU. Co-expression of nejScer\UAS.T:SV5\V5 partially suppresses the increased cell death and aberrant axonal targeting observed in third instar eye discs of flies expressing Hsap\APPAβ42.Scer\UAS.cUa under the control of Scer\GAL4GMR.PU.

Complementation and Rescue Data
Comments

Expression of nejScer\UAS.T:SV5\V5 rescues the mitosis phenotypes in eye discs resulting from Scer\GAL4hs.PB-driven nejdsRNA.Scer\UAS expression.
