Overexpression of ftScer\UAS.cMa under the control of Scer\GAL4Bx-MS1096 affect wing shape and alters posterior wing ridges. ftScer\UAS.cMa overexpression alters wing hair polarity in the proximal region as well as it also generates variable hair polarity changes in more distal regions of the wing.

Overexpression of ftScer\UAS.cMa under the control of Scer\GAL4salm-459.2 does not affect wing hair polarity.

Expression of ftScer\UAS.cMa under the control of Scer\GAL4en-e16E in the posterior compartment of the wing results in misorientation of wing hairs within the expression domain. These animals show defects in the directional preferences of growing microtubules in wing cells 24 hours after puparium formation; microtubule polarity in the proximal region of the wing is reversed, with a significant excess of plus-end proximal microtubules compared to wild type. The proximodistal alignment of microtubules becomes less strong compared to wild type.

Generalized overexpression of ftScer\UAS.cMa under the control of Scer\GAL4arm.PS has little effect on denticle belt polarity.

Overexpression of ftScer\UAS.cMa in the P compartment (under the control of Scer\GAL4en-e16E) causes 'whorly' polarity in the P compartment of the wing and tergites. Expression in larvae results, in some cases, in row 1 cells making clusters of bristles instead of rows of normally oriented denticles. Overexpression under the control of Scer\GAL4ptc-559.1 in the region of the denticles does not result in changes in polarity.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL in a wild-type background result in a reversal of cell polarity in wild-type cells anterior to the clone (hairs in front the clone point forwards).

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL in a ft12/ftunspecified background do not change the cell polarity of cells surrounding the clone.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are mutant for ftunspecified and are also expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL result in a reversal of cell polarity in cells anterior to the clone.

Following overexpression of ftScer\UAS.cMa, under the control of Scer\GAL4e22c, in the posterior wing compartment, cells flanking the DV boundary display wider apical circumferences when compared with those of the anterior wing compartment. Wing growth along the proximal-distal axis in the posterior compartment is truncated, as revealed by the close proximity of the anterior and posterior crossveins.

S2 cells that express ftScer\UAS.cMa, under the control of Scer\GAL4Act5C.PI, aggregate.

Expression of ftScer\UAS.cMa under the control of Scer\GAL4Act5C.PI causes mild perturbation of planar cell polarity in the wing and a foreshortening along the proximodistal axis of the wing blade as the anterior and posterior crossveins are abnormally close.

ftScer\UAS.cMa; Scer\GAL4Act5C transfected S2 cells do not aggregate.

When ftScer\UAS.cMa is overexpressed under the control of Scer\GAL4Bx-MS1096 in a pk30 mutant wing, the pk30 wing hair phenotype is modified to a more distal polarity in the anterior and distal posterior regions.

Overexpression of ftScer\UAS.cMa under the control of Scer\GAL4salm-459.2 in a pk30 mutant wings results in changes in wing hair polarity (relative to the wild-type pattern) outside of the main Scer\GAL4salm-459.2 expression domain.

dsUAO71 ft15 double mutant clones in the dorsal adult abdomen where the clones are also expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL cause repolarisation in front of the clones, so that hairs point away from the clone (64% of clones repolarise with a maximum range of seven cells). Co-expression of fjScer\UAS.cZa within these clones strengthens the repolarisation (95% of clones repolarise with a maximum range of seven cells).

The planar polarity defects seen in the wing in animals expressing ftScer\UAS.cMa under the control of Scer\GAL4en-e16E is significantly suppressed by co-expression of par-1dsRNA.Scer\UAS.cZa.

The reversal of cell polarity that is seen in cells anterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL is still seen if the clones are induced in a stan- background (the cells within the clone are stanE59/stanE59 while the cells surrounding the clone are stan3/stanE59).

The reversal of cell polarity that is seen in cells anterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL is still seen if the clones are induced in a fzunspecified background.

The reversal of cell polarity that is seen in cells anterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL is still seen if the clones are induced in a fzunspecified stan- background (cells within the clone are stanE59/stanE59 while the cells surrounding the clone are stan3/stanE59).

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are mutant for fzunspecified and are also expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL result in reversal of cell polarity in cells posterior to the clone when the clone is at the front of the anterior compartment (behaving like fz- clones) and result in reversal of cell polarity in cells anterior to the clone when the clone is at the back of the anterior compartment (behaving like clones expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL).

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) that are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL in a dsunspecified background do not change the cell polarity of cells surrounding the clone.

Clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are doubly mutant for dsunspecified and ftunspecified and are also expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL can reverse the cell polarity of cells anterior to the clone.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are co-expressing both ftScer\UAS.cMa and dsScer\UAS.cTa under the control of Scer\GAL4αTub84B.PL result in a reversal of cell polarity in cells anterior to the clone.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are co-expressing both ftScer\UAS.cMa and dsecto.Scer\UAS under the control of Scer\GAL4αTub84B.PL result in a reversal of cell polarity in cells anterior to the clone.

The reversal of cell polarity that is seen in cells anterior to clones in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are expressing ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL is enhanced if the clones are induced in a fjunspecified background compared to a wild-type background; the repolarisation occurs over a greater range in the fjunspecified background.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are co-expressing both fzScer\UAS.cUa and ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL and the clones have been induced in a dsunspecified background, result in reversal of cell polarity in cells anterior to the clone.

Clones of cells in the anterior compartment of the dorsal epidermis of the adult abdomen (in the tergite) where the clones are co-expressing both fzScer\UAS.cUa and ftScer\UAS.cMa under the control of Scer\GAL4αTub84B.PL and the clones have been induced in a ft12/ftunspecified background, result in reversal of cell polarity in cells anterior to the clone.

S2 cells that express ftScer\UAS.cMa and dsScer\UAS.cTa, under the control of Scer\GAL4Act5C.PI, aggregate, as do cells co-transfected with ftScer\UAS.cMa and dsΔICD.Scer\UAS.

S2 cells co-transfected with dsScer\UAS.cTa, ftScer\UAS.cMa and Scer\GAL4Act5C aggregate into clusters of about 50 cells. This aggregation occurs even if 2 groups of cells are transfected separately with dsScer\UAS.cTa; Scer\GAL4Act5C or ftScer\UAS.cMa; Scer\GAL4Act5C and then mixed. Formation of these aggregates is inhibited by the presence of EGTA. Co-transfection of fjScer\UAS.cZa with dsScer\UAS.cTa, ftScer\UAS.cMa and Scer\GAL4Act5C into S2 cells has no detectable effect on their aggregation, or the sensitivity of aggregates to agitation. Planar cell polarity defects in the posterior compartment of ftScer\UAS.cMa; Scer\GAL4en-e16E flies are weakly suppressed by ds05142/ds05142 and strongly suppressed by fjd1/fjd1.

Expression of ftScer\UAS.cMa under the control of Scer\GAL4Act.PU in ftG-rv/ft8 animals substantially improves hair planar cell polarity defects in the abdomen.

Expression of ftScer\UAS.cMa in the posterior wing pouch, driven by Scer\GAL4en-e16E, rescues the overgrowth phenotype of ftG-rv/ft8 mutants in this region, but does not rescue overgrowth in the anterior.

Expression of ftScer\UAS.cMa under the control of Scer\GAL4Act5C.PI rescues the lethality and wing disc overgrowth phenotype of ftG-rv/ft8 mutants and partially rescues the abdominal planar cell polarity defect.

ftScer\UAS.cMa; Scer\GAL4da.G32 rescues the pupal lethality of ftG-rv/ft8 and substantially rescues planar cell polarity and tarsal segment defects in these animals.
