Gcn5E333st/Df(3L)sex204 transheterozygous larvae present morphologically normal nephrocytes. In adult escapers, pericardial nephrocytes are frequently abnormally-shaped.

Gcn5E333st/Df(3L)sex204 transheterozygotes rescued to adulthood by the expression of Gcn5F304S.UAS.Tag:FLAG under the control of Scer\GAL4da.G32 are flightless and exhibit fully penetrant blistered wing and small, mildly rough eye phenotypes, and exhibit frequent leg morphology defects. They show cardiac defects: significant decreases in cardiac output, diastolic diameter and fractional shortening, and significant increases in heart period and arrhythmia index. They also show abnormally-shaped pericardial nephrocytes.

Gcn5E333st/Df(3L)sex204 transheterozygotes rescued to adulthood by the expression of Gcn5UAS.Tag:FLAG under the control of Scer\GAL4da.G32 also show some heart defects: significant decreases in cardiac output and diastolic diameter, but no significant changes in heart period, fractional shortening and arrhythmia index. They also show abnormally-shaped pericardial nephrocytes.

Photoreceptor organization is disrupted in PcafE333st eye discs and there is a defect in the R-cell projection pattern.

PcafE333st/PcafE333st and PcafE333st/PcafQ186st animals develop to wandering third instar, but do not form a puparium at the normal time and continue to wander for an additional 4-5 days. When they eventually stop moving, they fail to evert their spiracles and form abnormally long prepupae. These die with partial separation of the posterior part of the prepupa from the pupal case. PcafE333st/PcafE333st animals fail to form normal (for late third instar) sized early polytene chromosome puffs (2B, 74EF, and 75B) during their extended wandering third instar stage. Imaginal discs from PcafE333st homozygous third instar larvae are misshapen and severely reduced in size . PcafE333st/PcafC137T and PcafE333st/PcafΔT280-F285 animals form their puparium normally, but during pupation they fail to elongate their metathoracic legs correctly and to deposit some or all abdominal cuticle. Rare adult escapers have misshapen wings, rough eyes, and crooked and twisted metathoracic legs and die a few hours after eclosion. In PcafE333st/Df(3L)sex204 females rescued to adulthood by PcafScer\UAS.cCa or PcafΔBromo.Scer\UAS with Scer\GAL4da.G32, oogenesis is arrested at stage 5-6.

One copy of PcafE333st enhances the reduction in eclosion rate seen when Hsap\HTTQ93.ex1p.Scer\UAS is expressed under the control of Scer\GAL4elav-C155. These flies have a relative eclosion rate of 49.4%. PcafE333st/+ also enhances rhabdomere loss, with an average of 4.32 rhabdomeres per ommatidium.

Expressing Gcn5F304S.UAS.Tag:FLAG under the control of either Scer\GAL4da.G32 or Scer\GAL4αTub84B.PL partially rescues the full lethality of Gcn5E333st/Df(3L)sex204, giving rise to a few or to nearly half of the expected adult progeny, respectively.

PcafΔHAT.Scer\UAS; Scer\GAL4da.G32 fails to rescue the lethality or reduction in polytene chromosome acetylation due to PcafE333st/Df(3L)sex204. PcafΔAda.Scer\UAS; Scer\GAL4da.G32 rescues polytene chromosome acetylation, but not lethality do to PcafE333st/Df(3L)sex204.
