Expression of EBV\BZLF1GMR.PA results in dose-dependent eye phenotypes including rough eye, diminished pigmentation, and loss of ommatidia.

Flies carrying EBV\BZLF1GMR.PA show loss of all eye structures except the interommatidial bristles, which are disorganised and scattered throughout the thin cuticle that remains in place of the eye. EBV\BZLF1GMR.PA third larval instar eye discs show a significantly smaller number of cells which incorporate BrdU compared to wild-type discs and there is an increase in cell death in the discs compared to wild type. Flies carrying a weakly expressing P{GMR-BZLF1.A} line show a dose sensitive eye phenotype, with flies carrying one copy having a moderate eye phenotype with some eye tissue (but no recognisable ommatidia) and flies having two copies lacking all eye structures except the interommatidial bristles.

TorΔP/+ partially enhances the eye-phenotype associated with EBV\BRLF1GMR.PA-expression.

Tork17004/+ partially enhances the eye-phenotype associated with EBV\BRLF1GMR.PA-expression.

While EBV\BZLF1GMR.PA expression alone does not significantly alter the number of mitotic cells in eye discs, expression of EBV\BZLF1GMR.PA reverses the EBV\BRLF1GMR.PA overproliferation, such that the number of mitotic cells is lower than in wild-type controls. The number of short hair-like bristles in the adult eye is also decreased. The eye color of the double-transgenic flies is very similar to that of single-transgenic EBV\BZLF1GMR.PA flies (orange), suggesting that pigment cells do not over-proliferate as in EBV\BRLF1GMR.PA-eyes. The double-transgenic flies show a complete loss of ommatidia, although the bristles remain.

The eye phenotype of flies carrying a weakly expressing P{GMR-BZLF1.A} line is unaffected by dapGMR.PdN, RbfGMR.PD, CycEAR95 or BacA\p35GMR.PH. The eye phenotype of flies carrying a weakly expressing P{GMR-BZLF1.A} line is enhanced by svspa-pol/+.
