Expression of hpoScer\UAS.cUa under the control of Scer\GAL4nub.PU results in a reduction in wing area compared to controls.
Expression of hpoScer\UAS.cUa under the control of Scer\GAL4GMR.PF results in extensive eye degeneration.
Overexpression of hpoScer\UAS.cUa in the GMR domain of the developing eye (under the control of Scer\GAL4GMR.PF) induces massive cell death without disrupting cell differentiation or patterning. In mid-pupal retina (around 24 hours after pupal formation), both photoreceptor and pigment cells continue to die precociously. In comparison, in wild-type pupal retinae cell death occurs prominently in the pigment cells, and around the retinal boundary. Compared with wild-type adult eyes, hpoScer\UAS.cUa overexpressing adult flies are arrested as pharates at room temperature, and at 18[o]C show small rough eyes. hpoScer\UAS.cUa overexpression causes apoptosis in all epithelial tissues, for example, in the wing disc short pulses of hpoScer\UAS.cUa expression using heat shock (Scer\GAL4hs.PB) causes apoptosis. In comparison, very few cells undergo apoptosis in wild-type wing discs. Overexpression of hpoScer\UAS.cUa in the eye, under the control of Scer\GAL4GMR.PF, results in a 'small eye' phenotype in the adult, caused by the induction of cell death.
Expression of hpoUAS.cUa under the control of Scer\GAL4dpp.PU causes undergrowth of the expressing region of the wing disc in the larva. Expression of hpoUAS.cUa under the control of Scer\GAL4nub.PU causes an extreme reduction in the size of the adult wing.
Expression of hpoScer\UAS.cUa in the germline, under the control of Scer\GAL4nos.UTR.T:Hsim\VP16, does not result in germ-cell over-proliferation, or any alteration of cell morphology or loss of fertility.
Expression of hpoScer\UAS.cUa under the control of Scer\GAL4nub-AC-62 results in a small wing phenotype.
Expression of hpoScer\UAS.cUa, under the control of Scer\GAL4GMR.PF induces apoptosis in the GMR expression domain, posterior to the morphogenetic furrow.
hpoScer\UAS.cUa; Scer\GAL4GMR.PF flies have severely reduced and mispatterned eyes. At midpupal stages, the developing retinas in these animals are disorganised with very reduced numbers of inter-ommatidial cells. Increased levels of cell death are seen in the eye discs of these animals at late third instar.
Overexpression of hpoScer\UAS.cUa, under the control of Scer\GAL4GMR.PF, during eye development induces apoptosis and results in small and rough eyes. Expression of hpoScer\UAS.cUa in the wing, driven by Scer\GAL4nub-AC-62, causes a reduction in tissue size.
Overexpression of hpoScer\UAS.cUa, under the regulation of Scer\GAL4C5 results in a large increase in apoptosis. Overexpression of hpoScer\UAS.cUa, under the regulation of Scer\GAL4C5 during imaginal disc development results in loss of eye, head, and wing tissues through the arrest of cell proliferation and the induction of apoptosis.

The reduction in wing area seen in flies expressing hpoScer\UAS.cUa under the control of Scer\GAL4nub.PU is not enhanced by co-expression of either RtGEFScer\UAS.T:Ivir\HA1 or GitScer\UAS.T:Hsap\MYC alone, but it is significantly enhanced by simultaneous co-expression of both RtGEFScer\UAS.T:Ivir\HA1 and GitScer\UAS.T:Hsap\MYC together. The strong reduction in wing area seen in flies co-expressing all of hpoScer\UAS.cUa, RtGEFScer\UAS.T:Ivir\HA1 and GitScer\UAS.T:Hsap\MYC under the control of Scer\GAL4nub.PU is unaffected if they are also carrying either PakKK101874 or Pak3GD8481.
Co-expression of hpoScer\UAS.cUa and ykiScer\UAS.cHa in eye-antennal discs, under the control of Scer\GAL4GMR.PF, completely rescues the small eye phenotype of hpo activation. Co-expression of Ncpro.Scer\UAS with hpoScer\UAS.cUa in the developing eye under the control of Scer\GAL4GMR.PF results in increased cell death, compared to either single mutant. These larvae develop into pharate adult stages with small eyes. Co-expression of Ncpro.C-A.Scer\UAS with hpoScer\UAS.cUa in the developing eye (both under the control of Scer\GAL4GMR.PF completely suppresses the cell death seen upon expression of hpoScer\UAS.cUa alone. Co-expression of ArkScer\UAS.T:Zzzz\His6,T:Hsap\MYC, Ncpro.Scer\UAS and hpoScer\UAS.cUa in the developing eye, under the control of Scer\GAL4GMR.PF induces massive apoptosis in the eye-antennal disc and adult. This phenotype appears stronger than controls as both photoreceptor and pigment cells are eventually eliminated in animals co-expressing hpoScer\UAS.cUa.
The small wing phenotype caused by expression of hpoScer\UAS.cUa under the control of Scer\GAL4nub-AC-62 is strongly suppressed by one copy of ykiDbo1, ykiDbo2 or ykiDbo3.
In eye-discs mosaic for ft8, the expression hpoScer\UAS.cUa under the control of Scer\GAL4GMR.PF induces apoptosis as in wild-type tissue.
The adult eye phenotype and the loss of interommmatidial cells seen in hpoScer\UAS.cUa; Scer\GAL4GMR.PF mid-pupal retinas is not suppressed by exBQ/exBQ or in exBQ Mer4 double homozygous clones, but is suppressed by wtsx1/wtsx1. Coexpression of banEP3622 with hpoScer\UAS.cUa, both driven by Scer\GAL4GMR.PF, significantly suppresses the eye defects and eliminates the apoptosis caused by hpo overexpression. Coexpression of banEP3622 with hpoScer\UAS.cUa in the wing, driven by Scer\GAL4nub-AC-62, suppresses the Scer\GAL4nub-AC-62>hpoScer\UAS.cUa small wing phenotype.
Coexpression of thScer\UAS.cHa and hpoScer\UAS.cUa, under the control of Scer\GAL4ptc-559.1, in wing discs leads to a folding of the epithelium in the ptc expression region. Clones in the wing disc that express hpoScer\UAS.cUa, under the control of Scer\GAL4tub, (rescued for apoptosis by expression of thScer\UAS.cHa) cause nuclei to move basally, inducing cells to change shape and causing a basally-directed folding of the epithelium.
Apoptosis induced by overexpression of hpoScer\UAS.cUa, under the regulation of Scer\GAL4unspecified is suppressed by coexpression of thunspecified.
