Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP in the pars intercerebralis (PI) under the control of Scer\GAL450y results in male flies with significantly increased fighting frequencies. A stronger phenotype is seen when Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP is expressed under the control of Scer\GAL4Ilp2.PR.
Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP in pk neurons, under the control of Scer\GAL4ppk.PG and Scer\GAL80ppk.1.0 fails to trigger acetic acid attraction in yeast-deprived females.
Scer\GAL4Tdc2.PC Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP male flies eat significantly more food ad lib than controls in a 24 hr period and the average number of feeding bouts is significantly increased. These phenotypes are also observed in the absence of Scer\GAL4Tdc2.PC, though at a lower level.
Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4dome-PG14 results in a significant decrease in triacylglycerol levels in adult flies compared to wild type.
Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP in the ASM group of cells (under the control of Scer\GAL4Tdc2.PC and Scer\GAL80αTub84B.PL) produces a decrease in sleep similar to that seen with expression in the all neurons under the control of Scer\GAL4Tdc2.PC. The sleep loss is approximately 320 minutes when Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP is expressed in multiple cells of the ASM group, but less when it is expressed in only one cell in this group. Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP in other areas of the brain and thorax does not result in a significant drop in sleep.
Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP in a subset of PI neurons under the control of Scer\GAL4Ilp2.PR results in an average decrease in daily sleep of 172 minutes, mainly from nighttime sleep.
Neuronal hyperactivation of mdIV neurons through expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4ppk.1.9 display severely premature intermediate surfacing transition (IST) behavior (emerging from the food transiently to to roam the food surface, prior to complete food exit) compared to wild type. The mutant larvae also display a premature shift in thermotactic preference behavior (wild type larvae change the way they respond to temperature changes when they go from feeding to wandering) compared to wild type. However, eventual food exit does not occur significantly later than in controls. The resulting adult flies are smaller in size than their wild type counterparts.
Flies expressing Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of either Scer\GAL4c673a or Scer\GAL4fru-GAL4 have a decrease in fat levels compared to control flies. The abdominal fat cells have either no fat droplets or only a few small ones. The food intake of these flies is not significantly different from that of controls. These flies show an increase in CO[[2]] emission compared to control flies.
Flies expressing Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4fru-GAL4 show massive autophagic vesicles in fat and malpighian tubule cells (these are not seen in control flies).
Flies expressing Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4dimm-929 show a dramatic increase in nighttime activity compared to controls. The addition of Scer\GAL80Pdf.PS almost completely rescues the abnormal nighttime activity in the early part of the night.

Flies in which Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP is expressed under the control of Scer\GAL4dimm-929 in the large ventral lateral neurons (l-LNvs) of the brain (Scer\FLP1Ecol\lexAop.cSa protein produced by expression of Ecol\lexAPdf.cSa.T:Hsim\VP16 has been used to remove Scer\GAL80Tub.PS function (and thus allow Scer\GAL4 expression) in a subset of the Ecol\lexAPdf.cSa.T:Hsim\VP16 expressing cells) show an increase in nighttime activity compared to controls, with the activity being highly correlated with the number of LNvs expressing Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP. Daytime activity is unaffected.
Flies expressing Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4R6 show no significant alteration in sleep compared to controls.
Flies expressing Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4cry.PE in the presence of Scer\GAL80Pdf.PS show increased sleep compared to controls, especially at night in light-dark conditions, and the phenotype is more obvious under constant darkness conditions.
Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Mef2.247.Switch (in the presence of RU486) causes a decrease in sleep.
Electroretinograms from Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP expressing flies (under the control of Scer\GAL4unspecified) exhibit large, slowly activating and inactivating depolarisations that are absent in the electroretinograms of control animals. Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP in NCCAP neurons, under the control of Scer\GAL4Ccap.PP, results in complete failure of wing expansion, and inhibits tanning at levels comparable with 2xShEKO.Scer\UAS.T:Avic\GFP-GL. These effects are not accompanied by pupal mortality. Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4dimm-929 does not have a significant effect on wing expansion or cuticle tanning.
