Expression of Prat2dsRNA.IR-AS.Scer\UAS under the control Scer\GAL4Act5C.PI results in varying degrees of lethality, depending on the P{UAS-Prat.IR-AS} line used, the copy number and the temperature at which the flies are raised. Expression of Prat2dsRNA.IR-AS.Scer\UAS under the control Scer\GAL4αTub84B.PL results in 50% of arrested pupae having a "cryptocephal" phenotype, consisting of head eversion defects, legs that are not fully elongated, an abdomen that lacks cuticle and segmentation and larval mouthparts still being attached to the imaginal head structure. This cryptocephal phenotype is also seen in animals expressing Prat2dsRNA.IR-AS.Scer\UAS under the control Scer\GAL4Act5C.PI. Rarely, animals expressing Prat2dsRNA.IR-AS.Scer\UAS under the control Scer\GAL4Act5C.PI have a "microcephal" phenotype, which results from partial head eversion. These pupae retain their larval mouthparts and do not have a fully differentiated abdomen. Surviving female adults expressing Prat2dsRNA.IR-AS.Scer\UAS under the control Scer\GAL4αTub84B.PL are fertile.

Co-expression of both Prat2dsRNA.IR-AS.Scer\UAS and PratdsRNA.IR-AS.Scer\UAS under the control of Scer\GAL4αTub84B.PL results in a higher degree of female lethality than that seen when either Prat2dsRNA.IR-AS.Scer\UAS or PratdsRNA.IR-AS.Scer\UAS is expressed alone under the control of Scer\GAL4αTub84B.PL. 27% of the double mutant animals arrest at a very early prepupal stage. Some of these arrested prepupae retain the thin and long shape typical of larvae, the anterior spiracle fails to evert and the gas bubble remains in the middle of the body. 34% of the double mutant animals arrest at prepupal stage P4(i) and have head eversion defects and a shrunken abdomen, and the gas bubble is in the posterior of the animal. 24% of the double mutant animals show head eversion, but the head is partially collapsed, resulting in a large cavity in the anterior of the pupa. 7% of the double mutant animals have a "cryptocephal" phenotype and 5% die as pharate adults.
