Scer\UAS regulatory sequences drive expression of a 729bp genomic fragment of Sp7 (which includes the last 3 introns) and a 425bp cDNA sequence of Sp7 arranged in opposite orientation. The construct is expected to produce a dsRNA composed of a 425bp hairpin linked by a 148 nucleotide loop.

Expression of Sp7[dsRNA.Scer\UAS] in crystal cells under the control of Scer\GAL4[lz-gal4] results in reduced hemolymph clot melanization.
Flies expressing Sp7dsRNA.Scer\UAS under the control of Scer\GAL4tub show a severe reduction in the amount of melanisation produced at a wound site (flies were pricked with a needle dipped in a concentrated mixture of E.coli and M.luteus) compared to wild-type flies. Spontaneous melanisation of the hemolymph (assayed by dissecting larvae and exposing them to air) is much lower in flies expressing Sp7dsRNA.Scer\UAS under the control of Scer\GAL4unspecified larvae than in wild type.
