At 25[o]C, 70% of flwG0172 larvae fail to successfully pupariate and of the 30% that do form pupae, essentially all arrest development prior to adult eclosion.

Mutant larvae are reduced and more variable in size compared to wild-type controls. The mutant larvae show reduced motility compared to wild type and frequent detachment of muscles is seen, particularly in the ventral intersegmental muscles.

More than 99% of mutants die before reaching adulthood, and the few escapers have curled or crumpled wings.

Dividing neuroblasts from flwG0172 homozygous larvae have tangled, unevenly condensed and lagging and bridged chromosomes during anaphase

Around 2% of flwG0172 mutant males survive. Survivors have severely crumpled wings.

The indirect flight muscles are completely absent in mutant adults.

flwG0172/+ enhances the shorter egg length phenotype seen in Pkn1T/Pkn1T or Pkn2T/Pkn2T germline clones.

stckwt.T:Zzzz\FLAG increases the frequency of male flwG0172 escapers from 3.1% to 22.5% at 18[o]C. The rescued males are fertile. stckwt.T:Zzzz\FLAG increases the small size of flwG0172 larvae, such that they are increased in size compared to wild type. The reduced motility of flwG0172 larvae is rescued by stckwt.T:Zzzz\FLAG, with the rescued larvae being significantly more motile than wild type. The detachment of ventral intersegmental muscles seen in flwG0172 larvae is partially rescued.

At 25[o]C, stckwt.T:Zzzz\FLAG fully rescues the larval lethality of flwG0172 mutants, but the pupal lethality is not rescued.

flwG0172 mutants carrying either stckQ38A.T:Zzzz\FLAG or stckD303V.T:Zzzz\FLAG arrest during the larval stages. The larvae are significantly smaller and significantly less motile than flwG0172 single mutant larvae. Expression of stckQ38A.T:Zzzz\FLAG does not rescue the detachment of ventral intersegmental muscles seen in flwG0172 larvae. Expression of stckD303V.T:Zzzz\FLAG slightly rescues the detachment of ventral intersegmental muscles seen in flwG0172 larvae.

flwG0172 ; stck3R-17 /+ animals show complete larval lethality (only a limited number of animals were of this genotype could be isolated due to a high rate of balancer breakdown in the required crosses).

Expression of ZyxScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4how-24B has no effect on either the rate of larval or pupal lethality of flwG0172 mutants.

Pupariation is blocked in flwG0172 ; rheaUbi.PB mutants, with complete larval lethality being seen.

The wing defects of flwG0172 escapers are suppressed by Drakdel.

The T3 leg defects seen in Drakdel and DrakKO mutants are suppressed by flwG0172.

The addition of Mlc2S66A,S67A does not suppress flw mutant phenotypes. The viability of flwG0172 mutants is much greater in a sqhA21.T:Zzzz\FLAG background than a sqhA20.T:Zzzz\FLAG background. The crumpled wing phenotype of flwG0172 flies is partially rescued by zipSu(flw)1, zipSu(flw)2 and zipSu(flw)3 and fully rescued by Tm1Su(flw)4.
