Expression of GLazScer\UAS.cWa, under the control of Scer\GAL4D42, Scer\GAL4Act5C, Scer\GAL4hs.PB or Scer\GAL4elav-C155 at 29oC does not have any significant effect on lifespan. Expression of GLazScer\UAS.cWa in the central nervous system and some muscles, under the control of Scer\GAL412.1 results in an 8% increase in lifespan. Expression of GLazScer\UAS.cWa in the central nervous system and some muscles, under the control of Scer\GAL4109(2)80 at 29oC results in an 23% increase in lifespan. Expression of GLazScer\UAS.cWa in the fat and some muscles, under the control of Scer\GAL4Lk6-DJ634 at 29oC results in an 16% increase in lifespan. Expression of GLazScer\UAS.cWa in the fat and some muscles, under the control of Scer\GAL4how-24B at 29oC results in an 28% increase in lifespan. Expression of GLazScer\UAS.cWa in the eye and salivary glands, under the control of Scer\GAL4GMR.PF at 29oC results in an 23% increase in lifespan. Flies carrying both GLazScer\UAS.cWa and Scer\GAL4109(2)80 at 29oC display a 12% increase in survival under hyperoxia compared to the Scer\GAL4109(2)80 driver control. After 4 days in hyperoxia, flies carrying both GLazScer\UAS.cWa and Scer\GAL4109(2)80 at 29oC display both vertical and horizontal locomotor performances that are much improved compared to controls. No differences are apparent under normoxia. Overexpression of GLazScer\UAS.cWa under the control of Scer\GAL4109(2)80 leads to a 60% increase in lifespan under wet starvation and a 30% increase under dry starvation. There is no significant difference in dry weight, protein content, or fat content between GLazScer\UAS.cWa Scer\GAL4109(2)80 flies and controls. Flies overexpressing GLazScer\UAS.cWa under the control of Scer\GAL4109(2)80 that are subjected to hypoxia in 100% N2 for 30 minutes, then transferred back to a normal environment are much more active than control flies. One hour after recovery, approximately 75% of the overexpression flies climb up to the top third of the vial, while only 10-15% of control flies manage the same. This difference is still evident 2 days later. There is no difference in climbing ability before hypoxia exposure.
