Flies expressing Vmat[Scer\UAS.Ex.T:Ivir\HA1] under the control of Scer\GAL4[Ddc.PL] show a marked increase in basal locomotor behaviour compared to controls. This increase in locomotion is most apparent during daylight periods when flies are normally active. Flies expressing Vmat[Scer\UAS.Ex.T:Ivir\HA1] under the control of Scer\GAL4[Ddc.PL] or Scer\GAL4[elav.PLu] show a marked increase in the number of grooming episodes compared to controls. Flies expressing two copies of Vmat[Scer\UAS.Ex.T:Ivir\HA1] under the control of two copies of Scer\GAL4[Ddc.PL] show reduced climbing in a negative geotaxis assay. Flies expressing Vmat[Scer\UAS.Ex.T:Ivir\HA1] under the control of Scer\GAL4[Ddc.PL] show an increase in courtship activity, including wing song and the pursuit of female flies by the males. However, individual male-female pairs show a relatively low rate of sustained copulation; although occasional attempts are observed, they last only a few seconds and are terminated by the males dismounting and are often followed by the resumption of courtship behavior. The fertility of the mutant flies is decreased compared to wild type. Expression of Vmat[Scer\UAS.Ex.T:Ivir\HA1] under the control of Scer\GAL4[Ddc.PL] decreases behavioural sensitivity to cocaine.
