Flies that express Mnn1Scer\UAS.cCa under the control of Scer\GAL4how-24B, Scer\GAL4elav-C155, Scer\GAL4da.G32, Scer\GAL469B, or Scer\GAL4OK6 die at stage P14 and show deletion of distal elements of the proboscis and a melanotic mass at that location. The melanotic mass is visible as early as stage P7.

About 5% of flies expressing Mnn1Scer\UAS.cCa under the control of Scer\GAL4how-24B survive to eclose and about 25% of those expressing Mnn1Scer\UAS.cCa under the control of Scer\GAL469B. These eclosed files show a melanotic mass at the anterior proboscis, failed wing inflation and die after 2-3 days.

Flies that express Mnn1Scer\UAS.cCa under the control of Scer\GAL469B occasionally show a cleft thorax phenotype. 100% of flies that express Mnn1Scer\UAS.cCa under the control of Scer\GAL4pnr-MD237 show a dorsal cleft along the entire thorax with disrupted chaetae orientation. The expressivity of this phenotype increases with the number of copies of Mnn1Scer\UAS.cCa that are expressed.

Expression of Mnn1Scer\UAS.cCa under the control of Scer\GAL4ey.PH does not cause an eye phenotype.

Expression of Mnn1Scer\UAS.cCa under the control of Scer\GAL4pnr-MD237 in a Jra1/+ background results in an enhanced thoracic cleft compared to Mnn1Scer\UAS.cCa expression in a wild-type background.

The cleft thorax phenotype of flies that express either Mnn1Scer\UAS.cCa or JrabZIP.Scer\UAS under the control of Scer\GAL4pnr-MD237 is enhanced when Mnn1Scer\UAS.cCa and JrabZIP.Scer\UAS are expressed together by Scer\GAL4pnr-MD237. Further, flies that coexpress Mnn1Scer\UAS.cCa and JrabZIP.Scer\UAS under the control of Scer\GAL4how-24B are lethal at pupal stage P7, which is earlier than the pharate adult lethality of Scer\GAL4how-24B>Mnn1Scer\UAS.cCa flies.

Coexpression of Mnn1Scer\UAS.cCa and JraAsp.B.Scer\UAS under the control of Scer\GAL4pnr-MD237 results in 1-5% of flies escaping the JraAsp.B.Scer\UAS lethality to eclose. Viable flies have a severe cleft thorax. Likewise, the prepupal lethality of Scer\GAL4ey.PH>JraAsp.B.Scer\UAS flies is partially suppressed when Mnn1Scer\UAS.cCa is coexpressed.

A kay1/+ background has no effect on the Scer\GAL4pnr-MD237>Mnn1Scer\UAS.cCa phenotype.

Coexpression of kayScer\UAS.cEa and Mnn1Scer\UAS.cCa under the control of Scer\GAL4pnr-MD237 results in an enhancement of the cleft thorax phenotypes seen when either is expressed alone.

Coexpression of Mnn1Scer\UAS.cCa with kaybZip.Scer\UAS, under the control of Scer\GAL4ey.PH, results in complete suppression of the Scer\GAL4ey.PH>kaybZip.Scer\UAS rough eye phenotype.

Coexpression of Mnn1Scer\UAS.cCa and JraScer\UAS.cEa under the control of either Scer\GAL4pnr-MD237, Scer\GAL4ey.PH or Scer\GAL4how-24B results in the same phenotypes as expression of Mnn1Scer\UAS.cCa alone.
