UASt regulatory sequences drive expression of an inverted repeat.
Adults expressing TTLL3BGD7234 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
50% of embryos expressing TTLL3BGD7234 under the control of Scer\GAL4da.G32 do not survive to adulthood.
Almost all adult males expressing TTLL3BGD7234 under the control of Scer\GAL4da.G32 are sterile, with only 2.4% being able to generate offspring. Preindividualisation sperm cysts of mutant males appear similar to those of wild type. However, most mature cysts lack axoneme structures in the mutant males, while the mitochondrial derivatives are still present. In some cases, disorganised axonemes composed of singlet microtubules are seen. Rarely, cysts containing axonemes but with reduced or absent mitochondria are seen. Actin cones are scattered along the sperm tails.
Expression of TTLL3BGD7234 under the control of Scer\GAL4da.G32 in the presence of Dcr-2Scer\UAS.cDa to enhance the transgenic RNAi effect results in 100% lethality.
