Expression of BaceGD5366 under the control of Scer\GAL4αTub84B.PL results in complete lethality during the late-larval/early pupal stage.

Newly eclosed adults expressing BaceGD5366 under the control of Scer\GAL4Appl.G1a do not show a detectable mutant phenotype in the lamina. However, 100% of 4 week old flies have vacuoles in the lamina cortex and in the central brain.

Adults expressing BaceGD5366 under the control of Scer\GAL4GMR.PU show progressive degeneration of the lamina, with vacuolisation in the lamina cortex increasing over time. Although vacuolisation is present along the entire lamina cortex of 28 day old flies, vacuoles are not detected in the lamina neuropil or other brain areas. At 24-32 hours after eclosion, the first defects in the lamina cortex are detected close to the basement membrane. In 3 day old flies, gaps are seen in the row of glial cells in the subretinal layer, while the row of satellite and epithelial glia appear intact. Only a few glial cells are detectable in the subretinal layer in 3 week old flies and most of the remaining glia in the lamina cortex appear to be satellite glia.

Adults expressing BaceGD5366 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Co-expression of either Appls.Scer\UAS.T:Ivir\HA1 or AppldAICD.Scer\UAS.T:Ivir\HA1 has no effect on the lamina degeneration phenotype seen in 28 day old flies expressing BaceGD5366 under the control of Scer\GAL4GMR.PU.
